Let's test some new isolates! (Log) * 3

Well, some of them are clone isolates, the others are 4th and 5th generation transfers from spores that LOOK pretty isolated, you know how that goes. :smirk: Only the clone has ever been fruited, so everybody's in for a surprise with the others!

Cultures currently on agar:
  • B+
  • Colombian Rust
  • Clone (forgot what it was originally named, doesn't matter anyway)
  • Pan cam (sandoze)

There's also a clean LC of the clone as well as Rust available.

Here's what's prepped:


Above: 13 quarts of wheat berries (one plastic container as well)
Not pictured are another 8 quarts divided among 3 spawnbags.


Above: some grain masters inoculated with B+ (small containers) and the clone (bigger ones)


Above: Rust master jars, inoculated with an agar wedge about 2 weeks ago.
Below: B+ master jars, same story. Almost finished.



Bonus: pic of the clone fruiting in a micro-mono (bread loaf pan)



I'm fairly certain that the clone will guarantee pin porn for all, but I also want to see some red spore action. I'm thinking about inoculating the grain with 1/2 Rust and dividing the others between B+ and clone.

I'm undecided on how I want to inoculate it. I like agar wedges for the cleanliness, although the LC is an option as well and I prepped the grain a little dry in anticipation of it. The finished jars are practically begging for G2G, but I'm not sure I'm comfortable enough doing it in my small SAB to be confident I'll be able to see the project through that way, hah. The plastic containers are a bit easier to handle in there but it'll be a bit before they're ready to work with.

I could go ahead and spawn the finished jars so everybody can see some fruits soon, also. Any votes for that? Any other ideas for this project as a whole or anything in particular that anybody'd like to see? Hopefully there will be enough spawn to be able to have a little fun experimenting with it soon!

I'd like to see a constant update on a wedge, they're far and few in comparison to the MS crapshoot and G2G inoculation.

Whatever you decide to do, keep us posted!

I like that idea, I've really been liking wedges for inoculating, I used to assume they were slow, but honestly with a shake they're not bad at all. I think I'll inoculate 6 jars with wedges of the clone isolate and 6 with the LC (made from a wedge a few weeks ago) and see if there are any differences in how they behave, although I expect they'll turn out similarly.

The rest will probably all be Rust from wedges.

2 week update:

First, LC jars.


Next, agar jars.


Hmmm... something isn't right here...

Damn! Never seen trich in a spawn jar before. No idea how this happened, guess I picked it up during the transfer. Oh well, the other 5 jars are alright.

Let's take a closer look at how they compare:

The LC jars aren't all totally even, colonization-wise, although they appear to have more mycelium so far. The agar ones look pretty consistent.  They all got a shake after this, regardless. I'm guessing there won't be too much difference in time til they finish. When I do the initial agar inoculation, I cut a piece about the size of a fingernail, then score it a couple times and spear the slices to drop them in the jar, then reseal and rattle it a little to separate them and get a few starting points.

Bonus pics: Had a jar of spawn from a previous batch that smelled a little sweet, so I dumped it in a gallon jug and cased it with peat to see how it'd do. Far from ideal, but it seems to not be entirely wasted! I suppose you'd call this a "monojug", heh.

dude, agar to grain is the only way u should go.  no question.

lc to grain, vs one agar wedge to grain, the lc can win a lot. but why risk it?

Update:
3/4 rust jars from wedges were spawned to a medium box with sterilized coir/verm. I have no idea what I did with the 4th one, I think it's in one of the many unlabeled trays around here.




At first, I was like :confused: at where all the pins were, then I noticed this:



Yep, looks like 99% of this one's gonna be sidepins. I know a liner would prevent this but I like watching tubs fruit on all sides.

Anyway, I spawned 4 jars of the LC spawn and 3 jars of the agar spawn into another medium tub and they're both speeding along nicely.





I put one of the B+ jars in an itty bitty tub and used the other two for playing around with.

Oh, that reminds me, I didn't post an update on Monojug. I ended up taking the lid off and just putting it in my greenhouse. It, too, has elected to ignore the casing layer and go straight for the side pins directly off the grain. I could probably coax side pins out of an outdoor bed, honestly, I guess it's my specialty.



As for the BRF bricks, I've done a lot of them with sawdust/bran and oysters but these are the first 3 cube ones I've fruited and I'm pretty pleased with them! I basically treated them like cakes and cut the tops off after they'd consolidated so I could dunk them in the bag. I just left the bags on for humidity and so I could dunk them again easily, I thought about rolling one in verm but didn't get around to it. I put them outside sometimes for an hour or two of sun and rain when it's available.

I did get a another, tall tub so I'll try a liner with it, the second run of Rust spawn is done colonizing and will be used today or tomorrow.

Hah, right you are! Therefore, all potential fruiting containers (and space) ARE fruiting containers!


Update on the rust box: I was complaining about few top pins and low-and-behold, many more of them appeared today!



Spawned another 4 quarts of Rust into a 27 qt box and two smaller ones... soon my living space will be more container than habitable area.

Rust box is on its second flush. Not much new to see there. Turned out to be totally sporeless! That's kind of annoying since I really wanted prints, but oh well.

The two big tubs from LC caught trich and were buried. Not sure if I'm ready to blame the LC since it was started with a clean wedge and used successfully for at least half a dozen dozen tubs and all three BRF bags prior to this. I'm testing it on a couple pints and half pints to check so we'll see. Oh well. Agar tub's doing okay, though!


I really like this greenhouse, also. I like monos too but I very much like the greenhouse since I can fill it with random containers and not have to deal with a buncha perlite.

I love these bags. They took a long time to colonize but pin with immense fury.



One of the small tubs I buried a while back grew into this nice little outdoor patch.


Picked the big ones and left the small ones... 245 g wet. I love nature. This monster was 50 g by himself. I'll save him for a special occasion!

Nothing worse than having your LC test jar run smooth, only to find it contamed in between. Has happened to me. As for your sporeless rusties, I found that my first flush with them was also very light on spore production and I got better results from later flushes. This actually is not that uncommon and I have had it happen with other varieties as well. So try printing lots on your second flush.

Anyway nice fruits man :super:
