Whats the deal with FastFred's Media Cookbook?

http://www.shroomery.org/9393/FastFreds-Media-Cookbook

Is there more to it? Cuz I can't seem to find the info.

I have some agar-agar and want to try a no pour recipe.

Want to try a no-pour method?  Great; I love them.  Forget that old cookbook--what you really want is 2% agar by weight, 4-6% starch (any source will do) by weight, and a TINY (0.5%ish) amount of simple sugars, also by weight.  Heat, mix, pour into containers and then PC.  If you're looking for more in-depth instructions, I highly recommend Pasty's no-pour tek.  The addition of food coloring is especially helpful.

Ok cool man. So I just heat it to a gentle boil well stirring and PC for 30 mins once its cooled? What exactly is a simple sugar?

Yep.  Heat to mix, pour (or spoon) into containers, and PC for 30 minutes once it's cooled back down to room temp.

A simple sugar is another name for a monosaccharide.  Glucose and fructose are the most easily available ones, and you can get them in the form of corn syrup, agave nectar or even honey (although honey isn't recommended for germinating spores).

Ok, so you don't want 3-4% nutrient content in your agar. It'd be 3% tops, I usually shoot for 2%. Agar doesn't have to be at 2% either, it depends on what kind of agar you use and its' gelling strength.

The recipe I use is 15-20 g of agar agar and 20 g of extra-light malt extract per L of water.

FastFred's cookbook is one of the best out there. It's not a guide on how to pour and do agar though, it's simply recipes you mix and pour.

No-pour agar fucking sucks. A jar is NOT a petri, it's much harder to get your scalpel in there and get a tiny piece without getting your hand over it. They collect condensation like no other. The lids can be a bitch to get off and on.

Just get petris.

I do have petrie but how do I sterilize the agar in there?

You don't. You sterilize agar in another bottle then pour it into your sterile petris in a SAB or in front of a hood.

No, otherwise you get melted petris.

When I did a test on this, I got my best results at 6% starch, believe it or not.  I absolutely agree that it doesn't have to be 2%, either, but I prefer a stiff gel.

I'm sure you did. I bet it was much thicker but slower. The tests have been done over and over. You generally want 2-3% nutrient content for fastest growth. Maybe higher for slants.

I mean I tested the same isolate on 0%, 2%, 4%, 6% and 8% starch to see which would grow the fastest.  There wasn't a hell of a lot of variation, but 6% was the winner.

:whathesaid: I find that more nutes and softer agar is better for spore germination, less nutes and stiffer agar better for isolation and transfers. As for no pour agar, I find it much easier to work with in a SAB than pouring plates. Its all about personal preference and no pour has worked well for many people. Every grow pic I have posted save for a handful was started with no pour.

I imagine lower nutrition would be more conducive to...rhizomorphic growth???

I've read that many times, but it hasn't been my experience.

Lower nutrient content is more conducive to faster growth because they myc is searching for food. Yeah it'll be more rhizo too but that doesn't matter.

Try it with different isolates. Yours just happens to like 6%, that's not a general rule.
