Excellent Spore Rehydration Media
    #639506 - 05/21/02 04:34 PM (22 years, 8 months ago)

Just to pass along the results of a pseudo-test.

Two test tubes of L-broth (10 mL broth) were each innoculated with a large innoculation loop worth of spores that were sitting around for approx 8 months. Tubes were placed at 72-75 F; 48 hrs later cloudy mycellial network was visible surrounding/intermixing the spores.

L-broth: 1% peptone, 0.5% yeast nutrient, 0.5% NaCl, pH = 7.2 (contact your local science supplier)

Experiment was done because no growth was seen from a mycellium xfer to honey water (pH=?) done over a month prior.

I usually have very good results with honey water tec but I have seen others have problems. What was ratio of honey to water...I wonder if that may be reason for so many failures. I just use about a teaspoon to a cup of water then I microwave for 5 mins.
I did some 1/2 pint jars aout 2 weeks ago with 1/3 cup chunky BRF, 2/3 cup vermiculite, and 1/3 cup of 4 day old honey water culture. mixed the jars up dry and did em in 300F oven for 1 hr let em cool and just dumped culture in, closed up and shook like hell. They colonized in 5 days at 80F. No contams. I'm not suggesting this as a way of growing jars but it worked.

I'm never SURE of anything
