Very cool, I had a few clones I put on agar, but instead of staying white and growing into the agar, the tissue turned green and sporalated.  I threw a few away because all they were growing was bacteria.  One plate is left.  I'm not sure if its going to contam or if the spores will germinate. I enjoy this species as an edible, so hopefully it will stay viable.

This is after 2 MONTHS of growth! Wonder if I'll ever get as far as fruiting this?

It took a long time for these to get fuzzy, I threw 4 plates away because of green or bacteria and 3 had some nice growth.

Terry, that looks very good, when I've tried to grow this, it was never as symmetrical as yours, and the agar stained dark brown pretty quickly. It did however grow very slowly. Maybe it likes a different medium or ph.

I used plain old MEA. I have 2 other plates growing that look similar. Though one has a notch in it where I took samples for slants. The slants had similar striation rings surrounding the inoculation square.

Nice to hear that someone else has tried this. It's reassuring to hear from another source that this growth is normal for the species. I keep having doubts. :smile:

Extremely slow growth on a standard medium is common for Chlorophyllum rhacodes, brunneum and olivieri. They must belong to all those species which grow much better, when B-vitamins (thiamine, biotin, folic acid) and amino acids (peptone, yeast extract) are added.
Puffballs also have such a crippled metabolism.

What is the typical source for the P?

I got Y....the wrong sort, of course (active ;0), but I think that
I can still use it, but I have no peptone to try.

Sure, I assumed that a carbon source like glucose or maltose would be present.

Got my peptone from a fellow grower, who got it from a bodybuilder. These guys usually buy large amounts for their protein drinks.

I'll send you some. You only need 0.5 gm per 500 ml. I got mine from a lab supply house.

Try oat flake agar.

Stir 75 gr. oat flakes in 1 L very hot (200 F) deionized water for 10 minutes.

Then filter out the larger particles by pouring it through some mesh, save the broth and then add 25 gr. agar.

Beware, this medium will be less firm then other agar media, so extra agar has to be added to compensate.

This is my third glovebox Agar pouring session ever. (a few finished grows from MS rye/coir monotubs under my belt, but was not that happy with sporadic flushes and small fruits/yields. I actually had better results with the spent/contam'd grain outdoors... 

I am trying to isolate a Cube monoculture, and these guys are on their third agar plate now. Started off a grain of colonized Rye from MS syringe.  transfered some rizos from these plates to a weaker DME strength agar to try and see if it still shoots off rizomorphic growth or circular. (pictures are of 2nd transfer from grain) 


I am also trying to isolate a monoculture of Psilocybe galindoi (galindii) var. Georgia ATL#7. I have noc'd up two half pints of rye but nothing yet after 5-7 days. However, MS spore drops on agar germinated fast! at least it looks like healthy mycelium to me. These pics are first generation on agar from Multispore drop.


I have never grown sclerotia strains, and can some one chime in here? To isolate a good sclerotia producing substrain, should i just leave them be in the plate and wait for them to produce sclerotia there? or would doing transfers until monoculture appearance be the same thing.. i ask because i read on the site that you could "accidentally" isolate a fruiting substrain instead which i do not want right now.. either way i saved half to keep inside their first plate to hope for fruiting and then clone a stone, and the other half of plates i did transfers to new plates in hopes to isolate a monoculture.

Here is Reishi on agar from MS drop. Is it normal to for me to see growth the next day?! it was super fast colonizing the plate, just like it does to my rye jars..

It looks like you're using WAY too many spores if you're planning on doing ISOS.  You're making a lot of work for yourself.

Very clean. Is that a piece of mushroom or a piece of wood that the xylaria was growing on? Did you disinfect in some way before you put it on agar?

It looks like you're using WAY too many spores if you're planning on doing ISOS.  You're making a lot of work for yourself.



well i only used 1 Drop for each plate, the minimal i could get out of the syringe. im pretty sure it is because i tilted the plate while moving them and the spores spread out? either way, im gunna have to select something for the next transfer, might as well be from one of those small islands. (we are talking about the Reishi plates right, because that is why i almost thought it was a white contam, because there were so many small dots...)

If you spread the spores out like this, they have less chance of fertilizing each other to make a diversity of strains. Also, you won't be able to see clear radial sectoring because they don't all start growing from the center.

Oh, and one of those small perfectly circular islands is probably growth from a single spore, so it will be infertile.
