Isolating On Agar and Sectoring (with pics) * 1

I am just sharing some cool photos of my latest strain isolation work on Agar and maybe gets some input from some more experienced folks.

The photos are taken from the bottom while the dish is be held up against natural lighting.

Species is P. cubensis. I have done three transfers starting from MS with the most rhizomorphic, rigorous, and fastest mycelia. From the sectors on these two dishes I plan to pull close to 12 isolates and fruit them independently.

How many sectors can you actually make out on number #3?

Can you point out any broad sectors and segregating lines to the next? For example on #1 from about 8 O'Clock to 11 O'Clock, looks like one broad sector to me.

Feel free to make recommendations for my next transfer which will be this evening.

I think #3 has at least 8 sectors, but its hard to say because I think its kinda subjective.

Those are some of the best agar pictures I've ever seen. Nice work.

Did said transfers, a photo just for fun. I am trying to keep the project small, it wouldn't be difficult to end up with a 100 dishes doing agar work.



Another interesting topic; a little 'reusing plastic petri dish experiment'. About four days ago I was pouring some dishes and I just finished up and realized that I had enough agar left to pour another two or three dishes. Instead of grabbing new ones I decided to grab a few used plastic dishes. "what the hell, lets do a little experiment".

The used dishes had been washed with regular antibacterial dish soap and let dry. So I cracked em open and wiped the inside down with rubbing alcohol, they quickly dried in open air which had been ousted. Just for shits a giggles I poured the agar in open air as well. So four days later, no growth on any of the three dishes.

Seems to me if I was a little more carful and cleaned them and poured them in the box I could start recycling my dishes unless I am missing something.

Nikon D80, the lens isn't the best for close ups but it does the job. When it comes to DSLR's, more importantly is what lens you are using. I plan on purchasing another lens soon. Especially with the season coming I can take some better photos when I am out hunting. I took this photo of a Conocybe the other day. Beautiful mushroom under a wild berry tree. 



I digress, back to Agar. :wink:

It's coming along! I did another transfer session the other day, so now I am currently waiting on further growth to see how the sectors pan out. It shouldn't be long and I will be fruiting them to see if I can find a performer. In addition, I have a MS pan of the same strain fruiting and I will be taking multiple clones from that for a few different projects. Some hopefully to be preforming isolates for a future breeding project I have in mind.

I did lose one of the reused dishes that I talked about due to contams. I can get away with reusing them if I do everything right but even then, they just don't seem as clean and tend to build a little more condensation. I just recieved more in the mail, so I'm good to go.

This is one of those hobbies that I wish I had more time to do.. Working full time I have to limit the amount of time I can spend on it and size of projects.

Here is an update. A photo of one of the dishes front and back. They all look pretty much the same.  This is the 5th transfers from MS. Not sure why its no longer Rhizomorphic. Anyone?

What is the agar mix you are using currently?

The nutrient load in your agar mix can play a role in the visual
appearance of your mycelium. The less nutritious your agar is the more
likely that cube mycelium will exhibit rhizomorphic growth. The higher
the nutrient load the more likely you are to see cottony growth
even from a culture that has shown rhizomorphic mycelium on bulk
substrates or agar in the past.
