Question about Syringe to Petri dish
    #10571654 - 06/25/09 05:14 PM (12 years, 3 months ago)

Hi everyone! long time reader first time poster here, got a question about petri dishes.

well i decided to do my first grow so i ordered my supplies and syringes. since i work in a hospital and have access to the laboratory i picked up some petri dishes.(the kind with red medium)

SO my question is this:  can i place a couple of drops on the petri dish to grow mycelium and just inoculate jars of grain using the petri dish medium instead of the needle??

Will any mycelium even grow at all? because the petri dish has red medium instead of the potatoe/dextrose i see poeple use around here?

What is the ideal temp to incubate the petri dish ??

How long does it have to grow in order for it to be used to inoculate my grain jars?

Thank you for all your help!

The red stuff is usually sheep's blood...I don't think its the best medium for fungi, but it's reallyyyy good for growing bacteria so you might wanna stay away.  Go with PDA or Malt agar.

When you do get the correct agar, one drop will do.  If you only want to grow myc to inoculate shit then do a liquid culture, much easier and a lot more myc to work with.  If you want to work to isolate strains, then agar is right for you.  Nevertheless, an agar wedge is enough to inoculate a jar.  Incubate at room temp.

Search Agar TEKS and you will find a write up on Malt Extract Agar that I did, as well as a PDA written by hamloaf among others. As stated above the red medium you have is most likely blood based and for bacterial/viral cultures. Agar is not all that hard to work with. If I can do it with my makeshift mycology lab anyone can. Just be sure that your sterile procedure is upto par and you should be fine.
