Oasis of Growth in a Desert of Contamination... [Currently-> Pinning/Harvesting First Flush]
    #11376967 -

TLDR: Apartment is far from ideal
1) Apartment (despite my cleaning it regularly) is Dirty and Dusty. I spray raid often but still have little sugar ants that are coming in now that cold weather is here. I also have a cat who gets into everything (as well as unavoidable litter dust in the air)
2) Wildly fluctuating temperatures due to a large glass sliding door with terrible insulation and seals (70-85f in a given day)
3) No Pressure cooker

That being said, I am starting my first grow. I am attempting to deal with each of my issues thusly:

1) Keeping the colonizing jars in a sterile bucket covered with a towel on which I will liberally spray lysol daily. I also plan on keeping the aluminum squares on top of my jars after inoculation, as I seem to think that air exchange is not necessary during colonization.

2) Inside the bucket a large glass of water warmed with a fishtank-heater to keep the internal temperature of the bucket more constant (it has stayed between 77-80f over the last few days).

3) I plan on boiling my jars for 1.5 hours, I tested this method by filling the lower inch of a jar with white flour and water paste, sealing and cooking it as if I were sterilizing cake-jars, and letting it sit. It has been nearly a week and no sign of fungus has appeared.

My questions are - will my sterile bucket tek work in preventing my nasty apartment from infecting my jars? The humidity in the bucket is around 50%, but I have not yet kept the bucket covered. I lose about 1in of water from the glass every 2 days, will humidity buildup pose a problem?

Lastly, My glovebox is here, the gloves are not attached because I could not find any that were suitable. Instead I plan on washing my arms and hands thoroughly, using latex gloves, spraying the inside and gloves with lysol, and covering the front of the box (including my arms) with a towl drenched in lysol or alcohol during the inoculation process. Will this suffice?

I will be inoculating and sterilizing them tomorrow, my spores are coming in then (a syringe from a sponsor of this site).

Once your jars are inoculated either the dry verm layer for cakes, or your tyvek/filter lids for grains should act to keep contams out.
Usually any contams will come from the process leading up to incubation not the incubation itself. Humidity could contaminate your jars. Most people leave them at room temp to colonize and fewer folks are using incubators. If you have a closet or cabinet that will maintain even close to comfortable room temps, that would probably be better IMHO.
Hope it helps

I would really recomend that you do some more readng first because I don't quite understand your whole process. check out this link and the videos.

Sorry I better clarify. I was worried that keeping the inoculated jars simply in the open might contaminate them (because my air is so nasty). I constructed this bucket with the heater so they would be
A) Less likely to become contaminated.
B) Within the range for incubation.

gngsteroflove - thanks for clarifying, I had read on a few teks that handling the jars too much may allow contamination to pass through the dry vermiculite layer, and was worried that with my nasty environment even a little handling would ruin them. I will put them in my nightstand or something.

Does my glovebox seem sufficient? Is it really necessary for the gloves to be sealed to the box, or can I get away with what I have currently?

As far as your jars go, there is really not much reason to be handling them. Although you will probably want to look at them a lot, it is not something that you have to do a bunch. The layer of vermiculite on the top is very effective at reducing the chances for contamination. I would suggest that you keep them on a shelf or someplace at room temperature as long as it isn't an extreme temperature. Fluctuations between day and night time temps are fine. I wouldn't encourage you to use an incubator unless you have a very compelling reason for doing so.
What you really want to be concerned about is your sterile procedure while inoculating them. That is the critical part. Assuming that you get good sterilization of your cakes.
As far as your glove box goes, I have no doubt that you can make it work. Just do the best that you possibly can and you should be fine. remember that almost everyone has had problems and failures and that is part of the learning process. So learn to accept it if it happens then figure out what went wrong and where so that you can rectify it.
Good luck and let us know how you do.

Hey, I just re read your original post.

And you DO want to take the foil off of your jars after they are inoculated. they need GE (gas exchange). You can cover them with a coffee filter if you are worried.
If you use the incubator, just don't allow condensation to drip back down on top of your jars.

really take a good look at the first few videos when you click on this link. www.mushroomvideos.com
Watch the first few as many times as it takes. And go back to them again when you forget.

Thanks, I watched the videos and that clarified a little. I got my syringe today, mixed my substrate and am steam-sterilizing 8 1/2pint jars right now. Looking good...? Ill let it sit (after the hour of boiling) for about 6 hours while I go off to work before inoculation.

Mixin it up

adding dry layer on top + closing up jars

Foil on top...

...and into the pot

Glovebox shown above

I noticed some condensation on the tops of my lids when I took the foil off of them. Should I be worried? See below picture:

1 the lids should be upside down.
2 tape over the holes  Very important
3 90 boil

I am not using a grain tek, just the pf_tek - I assumed the 1in of dry verm was enough to keep contams out and taping over the holes was not necessary?

Like Gngstr said, the dry verm layer does very well at keeping out contams, so as long as your handling of them is contam free your jars will be fine during the few weeks of incubation. And you said you don't believe GE isn't important while colonizing...That's half-true, but still laughable. While it's not as important as in the fruiting phase, it's still needed by the myc for good solid growth (a buddy of mine actually did a trial about 3 yrs ago were he had 10 jars of treasure coast. he put 5 upside down on one of those wire-type closet shelves in his laundry room and the other 5 upright. the upside down jars colonized completely 6-8 days before the rest. why? b/c CO2 is heavier than oxygen thus, it escaped better and allowed for better GE in the upside down jars). That being said, if you're gonna put your jars upside down, flip them early and leave them alone. As far as temp goes, the temp in your place may vary widely but it's still in the acceptable range. Anything between 60-90f will do; while the rate of growth may be affected, they'll still grow.

it was just an example  eather way lids upside down

2 tape is nessisary to keep the water content right.  i havent got 6000 post just bullshiting people

No the tape is still very necessary. think of the dry verm layer as a bulletproof vest and the tape as a brick wall. if a contam (bullet) gets to you're bulletproof vest, it won't kill you, but you'll sure as hell feel it. I'd rather hide behind that brick wall with my vest on anyday.

yes if there is no tape it is not a DRY verm layer anymore wich defeats the perpouse

ok i have taped up the holes, when you say put the lids upside down do you mean flip the jars upside down? or should I have put the flat lid on backwards before I screwed the ring on top?

Tape goes on pre steam sterilization to keep condensation from the sterilizing pot from dripping into your jars.

The rubber side of the jar lid should be facing upwards.

I thought that was what the aluminum foil was for?

demonkinsin said:

What purpose does that serve? Dosent it make a better seal when the rubber is down-facing? It is too late for me to change it now, is it a big deal? That was never mentioned in any vid I watched or tek I read

here are the jars now, I used blue painters tape (all I have)- will that suffice?

O yes sorry your right about the foil. Benzos and beer awww lol.

But this is straight from PF Tek

When using two piece canning jar lids, the inner lid (with the rubber edges up) rests on the top of the jar and when the lid band is screwed off, the lid remains resting on the jar top. To make the lid and band act as one lid, place pieces of masking tape on the lid attaching the band to the lid. Then, the lid can be adjusted for air ventilation and looseness like an ordinary one piece jar lid.

Edit: Micropore tape is the accepted best but to my knowledge you should be ok with that tape.
