Agar questions

hey all i recently started my agar project and i had a few questions :

1. when you inoculate with a syringe do you just need a drip in a corner of plate( i got 3 section dishes ). when i knocked one up i squirted a lil bit of lc on it and the liquid ran across the whole section . or do i need to actually puncture the agar with syringe and inject into the agar?

2. how many dif types of inoculations is there for agar ?

3 . when done i just take micro pore all around dish label which section is which and throw it in same type incubator as jars ? ( i use a cardboard box with a heating pad under it .

4. is it better to pour agar thin ?

5. can i just clean used agar plates and put them in dish washer and wipe with iso when i am getting ready to pour again ?

6. i should be able to do agar fine in a glove box or do i need a flow hood ? is a flow hood better to have in general rather than glove box ?

7. if a plate becomes contaminated on one part in a 3 section dish is it very possible the other two parts will too ? if i get a dish contam can i just cut out a good clean looking piece of myc and do a transfer ?

i know some of these might sound stupid but i just want to be sure i looked up a few diff things i asked but didn't get a real exact answer .

anyone got any videos or pics of their inoculation process i would be glad to look at them . Ive seen rr videos they cover good amount of things very well

1. try to drip the liquid in the middle. dont puncture
2. spore print, spore solution, lc, agar transfer, tissue transfer im sure the list goes on
3. sounds good
4. I dont know tend to pour heavy personally
5. You will probaly get lots of contaims throw the plate.
6.It's better with a flow hood but a GB works it can just be tricky sometimes
7. Yes just transfer to a new plate

1) a drip- but in the center of a section, not a corner
2) spores, live mycelium, tissue, LC... at least those
3) Yep. You can put each one in a clean baggie if you like. Try not to move them or them or breathe on them too much.
4) dunno
5) Sterilize them.
6) Glove box at minimum, it'll work ok but you'll have some contams. You'll get better with practice, it's as much about technique as it is about equipment.
7) Likely- transfer healthy cultures away from contams ASAP.

edit- I had 3 and 4 switched.

I was kinda wondering about that doc.  before the edit.  I am pleased my answers were pretty close to yours but it seems you reuse plates

I also just started working with agar. Poured 20 dishes today and have a stack of dishes with lots of condensation in them. I think I didn't let the agar cool long enough after taking out of the pressure cooker (let it sit in PC for two hours). I also think I poured them a little thick which could've led to higher internal temps.  I say this as a point of caution when you pour, but I'd also like to know from the pros out there what I should do with these dishes. They were carefully covered with the original sleeve while still in the glovebox and 12hrs later condensation is the same. I'm concerned that the moisture will invite contams so...

Should I wait until it clears?
Noc them up as they are without worrying?
Toss them and start anew knowing that contams are very likely?

I squirted some LC on a dish.. of course it was no drip.. it was a squirt. The dish is a complete ugly mess of myc, but no contams :smile:

I will now be careful just to make a drop in the future..

mush mike i had condensation on some of mine as well but it seems that most say to pour at lower temps otherwise let the agar cool down alot then pour dishes .

they should be fine

He;s asking where in his local area.

Walk around the town look for science shops hobby store I was amazed how many shops carried stuff for this kinda work in my small town
