Be sure to make a few LC's of each, because LC's can be deceptive.

The white cloud you see in the liquid can be a contaminant!! :grin:

I look forward to that!  Like I said, I haven't grown anything since 2001 and I did the most ghetto PF tek imaginable.  I used drinking glasses instead of jars, boiled in a dutch oven for 45 minutes to "sterilize," covered with foil (no lids), used no glovebox, incubated in a cooler at room temp (maybe 72F) and so on and so forth.  I don't remember what strains I used, though I'm pretty sure at least 1 was B+.  Long story short, the glasses worked out fine (contam in 2 out of 10) and an old fishtank for a terrarium with a layer of perlite on the bottom was my growhouse.  After that, I just learned how to pick cyans and azures and gave up on growing.  All in all, I'm amazed anything grew.  A little older, a little wiser, and with a little dispensable income, I feel like I can do it up proper this time.

when I PC the jars, do the lids face seal side up or seal side down?

also, for incubation...fish tank heater in water in tub, jars in a tub within the larger?  is that an okay way to maintain temps?  any suggestions on maintaining incubation temp are welcome.

dont worry about incubating jars at a higher temp that info is outdated

It is outdated, but it does speed things up for me. Keep your jars near the ceiling if you can, much warmer up there. Remember that the mycelium generates heat too, so don't get your jars above 83 or so. It can be 5 degrees warmer in the jar itself.

In your case put the seal down. With Tyvek, I did seal down too. Just a matter of preference really...

Seal down, check.  I think I will still try to incubate in the high 70s if possible.  Submersible heaters are only $15 and if they speed things along, all the better.  Incubate in dark, or does it matter?

If you still got tinfoil over your lids from PCing be sure to remove it, colonization requires gas exchange.
Next time your out of the house stop and get some silicone, drill a hole through your lids and place a blob on both sides of it.
Works great, simply stick your syringe through it to inoculate and cut out any risks of contams.

I just read the self-healing tek and I'm going to go grab some silicone right now, before I PC the jars/WBS.

I'm also thinking I will just use this method of inoculation:

I still plan on doing everything I can to have a sterile room (lysol, no air flow, etc) but this seems like it makes the use of a glove box unnecessary.

I read that any 100% silicone will do.  Mine is clear, but 100% silicone.

It's good up to 400F as far as temps, so that should be okay, I'm assuming.

Cool just wanted you to be sure of the heat thing I use RTV but whatever will stand up to the heat.  Any silicone product will do the self healing thing

Alright - a new day.  I let the WBS soak overnight but I was having problems straining it: My colander is too porous.  So I threw it in a pillowcase, but quickly realized that it wasn't letting it drain enough and it was hard to rinse it.  Then I came up with an idea:  A mesh laundry basket!  I filled up the basket, which has mesh sides, then laid it on its side on top of the PC with a fan blowing on it (all in my shower, so it has a place to drain).  Now I'm getting close to the desired consistency.  No water drips out, but it still sticks to my hands a little much.  I'm going for "doesn't hardly stick to hand" wetness level.  I'm going to drain it another 30 minutes or so and see where we are.
