Cold agar, exposed petris, general sterile issues..

Hi all,

A few questions:

1. You open up a bag of petri dishes (say 20 per bag), lay them out in front of the flow hood and go to pour.  The agar is solid.  What do you do?  Trash the dishes because they are no longer sterile and you'd be annoyed as all hell if you spent time working with them later just to find out they got contam'd before they even were poured?  Is this a real concern.  The dishes were in front of the hood for a few minutes and placed back into the bag.  I hate throwing them away, but I also hate the uncertainty of them being dirty.. err. 

2. When working with things like slants and such, I have taken to wearing the full surgical garb.  Not cheap to buy tyvek sleeves, surgical masks, hair covers, etc.. How often do you trash these items (hair nets and gloves get trashed every time they come off for me) - specifically I am wondering about masks and sleeves.  Do you ever clean them - or just use them a handful of times and move on?

3. I have never poured a petri dish that didn't wind up with condensation.  I assume you haven't either.  I know people flip them, etc - but my question is, after pouring, do you put them back in their bag or let them "dry out" in the open air (after taping them up with micro-pore tape, that is)

Anybody else wonder how the pros do it?  This hobby is expensive enough without tossing $10 sleeves of petri dishes and $4.00 tyvek sleeves every time one sits down to work..

I know you can reuse the petri dishes by PCin them and I read somewhere on here there is a method to not getting condensation in your petries.  stack as you pore and put a empty on top??? something like that.  cant remember the thread.  Im sure if you searched about it you stumble across it.  as for the tyvek sleeves those are just for keeping your hair and skin from falling on your work area so I would bet they can be cleaned and reused.  if your talking about surgical masks they are washable and reusable.  the dust masks are useless in mycology and shouldnt be used cuz they dont stop anything but dust(if that) from entering your mouth and nose.

and when most people say "trash it" it usually means start over not really throw it away.  PCs sterilize alot of diff things if done properly.

There is plastic that can withstand a PC and plastic that can't.  Mine can't.  So, that isn't an option.

And potaonet, I'm in front of a laminar flowhood.

if you have any concerns pour your dishes and let them sit for 3 days before use. if no mold or bacteria shows up withing 3 days the plates are good to go.

Watch my video on pouring petri dishes.  By waiting until the agar has cooled enough to pour without heat protection for you hands, condensation will be minimal.  You should keep the dishes in a vertical stack, and after pouring, slip them back into the sleeve they came from.  When you remove and inoculate the dishes, wrap with parafilm and put in ziplock bags, three or four dishes to a sandwich size bag.  The ziplock bag provides an extra measure of protection.  Inflate the ziplock with air from your flowhood before sealing.

When you put the dishes in a stack, the condensation is re-absorbed back into the agar after a few days.

Pouring my Agar yesterday afternoon, I realised how badly designed my glovebox is. Argh, my kingdom for a flowhood.

I also waited until the MEA was cool enough to hold (too cool in fact, I managed to pour while lumps were forming), but still got a lot of condensation (stacked them as well).

Related topic, can the poly petris be cleaned out with bleach/alcohol/etc and be reused? I am getting some glass ones, but my order is taking forever to arrive.
