You're going to have to start off with a spawn, usually from jars. Liquid mycelium into a big container of manure wont do you much.
Use the mycelium and inject that into sterlized birdseed or rye grain. Let that colonize and use that as the spawn to innoculate your bulk substrate of manure. Make sure you pasturize your store bought manure as well.

You don't have too, but you'll have a much better chance at growing shrooms and not the green if you do. And do like Angryshroom said, use spawn, either a PF cake or grain (better than PF). And if you can, get some horse poo straight from a farm, you'll be happier than if you used the store bought. Good Luck.

Why will the liquid mycelium in the cow manure not work? If you are using something like karo, malt extract, or honey why can you not take a fully colonized jar and mix it in with a manure straw mixture or manure alone. You said it wouldn't do much but I guess I am just not following you

I don't understand your question. I did not say it wouldn't work, but I think you would have a better chance with bulk substrate, such as horse poo, with spawn such as colonised grain or a BRF cake, than you would with liquid mycelium. But thats just my opinion.

Its because you would need a lot of liquid mycelium, enough to change the water content of the manure, which would lead to slower colonization, which would lead to higher chance of contams. If you use spawn, such as grain or a PF cake, you have a lot of spawn without altering the water content.

First off, I think you have to do a little more reading. I really hate telling people this because its lame but anyways , im trying to help you out here with the simpliest answer:
Injecting liquid mycelium into a bulk substrate tub of manure would most likely not work. It COULD work under some good circumstances. However, its going to take quite a while to colonize, and that time to colonize is basically how you win or loose with manure. It will contaminate somewhat easy, especially if its overly-wet.
NOw you say something about some good substrates which you could colonize liquid mycelium with? Well you should really just inject that into some jars. Start small and then you take that spawn and mix it in with your bulk substrate like manure or straw or compost.
With store-bought manure you will still want to pasturize it. Just cook it in a boiling pot inside a laundry bag or pillow case for about an hour @ 170-180. Drain, squeeze, keep it moist but not wet.
Mix in your colonized grain or PF jars and incubate the manure until its colonized...
