grain problems
    #916665 - 09/29/02 12:51 PM (14 years, 2 months ago)

i have made 7 grain jars, sterilized them and after they were completely cool, ennoculated half of them with 1cc of acadian coast and other half with b+. it has been 5 days and there is no sign of germination in any of them and no contams either.

they have been incubating at 80 degrees
they all have a tyvek filter lid

does anyone have any idea as to why they didn't germinate?
this is my third round of grain jars and none of them have germinated.

Patience. Sometimes it takes longer than 5 days, to get germination.

Or, it could be that your grain was to dry, to wet or the spores are not viable.

One time this summer, I got a whole batch of non-viable spores. Not the vendors fault -- either. They got cooked in a outdoor mail box, in the sun & were subjected to very high temps, for 2 days.

i will give them another week.
6t how can you tell if the grain is too dry or wet? if anything it looks too dry.
i simmered it for 40 min then pc'ed it it looked sloppy wet when it came out but no it just looks swollen but definately not wet on the outside of the grain, maybe slightly waxy looking.
hopefully the spores are viable because the b+ was from sporeworks and the acadian was from tlg, they were sent at diff times too.

got a question about agar too, would it be a good idea to use the spore syringes on the agar? would'nt this just form a puddle on it?

I don't use grain much. I prefer a basic wild bird seed mix of red & white millet. I don't cook or simmer it. I simply soak overnight (12 up to 36 hours), then rinse, drain WELL, load & PC. Works every time.

Whatever seed or grain you use, Ideal moisture is between 40 & 60 %. With 50% = optimum.

I don't want to be pessimistic, but sometimes it simply doesn't work. The reason is not very important. I failed both with injecting up to 3cc of solution and quite a lot of spores directly on rye grain. Inversly, I also succeded from just an invisible scraping of spores.

The tossing: I'm waiting for four jars to innoculate form spores. (I use jars with the polyfill tek)
2 Puerto Ricans = 1. rye+straw, 2. birdseed.
2 Mazatapecs    = 1. rye+straw, 2. birdseed.
I tossed both birdseed jars on the second day: no sign of mycelium today (4th day).
I didn't toss the rye grain+straw jars: both started showing mycelium growth today.

I don't know if it's the tossing or the birdseed...

About three weeks ago, I injected 3cc of Pan. Trops in birdseed and rye grain. Only the birdseed jar worked (5 days later), the other one simply didn't want to. There were no tossing at all.

Thereby I mean what I said first: sometimes, it simply doesn't work. But there's only one solution: wait and see! If it doesn't work after a week and a half, either leave it (maybe it's going to work, after all!) and think about ordering another syringe or spore print (from a "local" FSR) or eat the grain... I like rye as much as my mushies do!

instead of starting over i have some honey water that is full of mycellium, atleast to my knowledge- it has many cottony slimy balls in the bottom. but i sterily sucked this into a sterile syringe and i am going to inject it into the grain jars, they do look like they are drying a little though, some grains look kinda shriveled
