Isolation necessary for clones grown on agar?

I was wondering if it would be benificial to transfer rhizormatic sections of clones that are grown on agar. One would think that if it is an isolate all the mycelium would grow identical. My eyes could be playing tricks on me, but it seems that there are both cottony and strandy sectors. Am I crazy, as all the mycelium from a clone is the same, go ahead and cut that sucker up and transfer to some grains, or should I isolate what could possibly be rhizormatic sectors? Thanks for any advice.

To be honest, I've never had problems transferring the cottony sectors from an isolate. They will grow out properly.

I thought that if the myc was sectoring it was not yet isolated. Were these isolates taken from multispore or a clone. Thanks.

Both, and I have seen what looks very similar to sectoring with isolates.

I don't know this answer for sure, I'm pretty much making some eduacated guesses but I've read the mushroom cultivator and it explains that there are certain monokaryon that are compatible with others and some that are not which will produce what they called illegitimate matings. I'm thinking that the cottony sectors might have something to do with this, and so I think that it would still be best to isolate the rhizomorphic mycelium. Somebody please correct me if I'm wrong.

I hope this helps. You should probably check out TMC even if I did manage to make sense above.

That is something I wonder about also. According to Stamets Ps. cubensis is ideally supposed to display rhizomorphic mycelia, but my zebra has some agar plates of an old Hawk's Eye B+ print which have pins forming on them with entirely cottony mycelium. ?? Not a rhizo in sight yet.

Because Cubensis fruits easily - even cottony mycelia will fruit. It takes a while to create an isolate that is purely rhizomorphic and is not sectoring - normally sectoring ALWAYS occurs with even the most rhizomorphic isolates because the nature of mushroom mycelia is that it is alwas changing. Ive seen highly rhizomorphic isolates produce nothing but cotton when isolated

Thanks for all the replies guys. It seems that there isn't a defenative anwser to my question. So I'm doing a little experminetation to try and find out for myself. I'll let everyone know how it goes.
