One way is:
You could do a tissue sample to liquid medium (ie.honeywater, dextrose water, light malt water).

Or you could make a mycelial slurry with the fruitbody.

I really doubt that the tissue sample to sterilised rye would work though.

I would cook some white rice so it is slightly overdone.
I would then put about a quarter cup cooked white rice in a pint widemouth.
After sterilizing for the customary time I'd let that jar cool .
Now, very carefully take a sample of tissue and transfer that to the jar of cooked ,sterilized white rice.
Thats the best way to proceed if you lack agar.

you can either use honey water or a mycelium slurry....
both works fine....with a personal preference to honey water...
or you can use peroxidated agar....

Whats your honeywater recipe? i have yet to try this
i tried, and fauiled myserably, with ME Liquid.

What about the slurry? how do you keep that sterile? whats your recipe with this?

Lizardking is the man to speak to about this... From my personal experiences you'll need to use h202 as liberally as possible.

you can prepare the slurry blending a clean piece of mushroom in sterile water and adding a couple cc's of H2O2 every 100cc of water.

Yeah i used a piece of mushroom tissue that was coffee grinded into a boil of h202. I then used a govebox to transfer it into malt/water and brf paste that was - as Club99 suggests about 2mls of h202 to every 100mls. It worked great!!
