Leaf cutting help :D

After reading allot of stuff yesterday about micro-propagation and other leaf cloning teks I have some questions.

Doing some salvia and basil leaf cuttings in like a million different ways. But I'm cheep and want to learn to use what I have. So I need hormones and I have a willow tree in my back yard and some urine in me lol just playing I wont use the urine.

OK so I have seen the yellow shoots of the willow tree is the best part to use and to soak them in water for 24hrs then use the water.

What is the ratio of water to branches?

Do I smash or crack the branches first?

Should I dip the branches in a 10% bleach for 10min or maybe a h202 just to clean off the dust then use distilled water 3 times to rinse with the bleach or just let the h2o2 break down?

Ill probably get some bean sprouts too and do the freeze thaw juice. They are only a buck so thats better then the time it would take to crack them lol.

OK now onto part two

Cytokinin is in the milk of a coconut.
I happen to have  one from last week.

So do I just crack it open and pour it into a clean jar and use that?

Should I boil it? pasteurize it?

Any other common plants with Cytokinin in it?

Its cool if there is no answers for some of my questions Its not the most common thing but any informed guesses are cool here too. Its just experiments.

I've been doing micro-propagation for about half a year now, I can't help you with biological sources of growth hormones since I've always just used the hormone in solution. You're going to want a pH meter or paper so you can adjust the pH to be slightly acidic especially if you're using impure sources of hormone - who knows how that will affect the media. In terms of sterilization you're going to want to rinse in alcohol first to remove some of the waxes and gunk ~1 minute, then 10% bleach w/ detergent solution [lowers the surface tension of the water] for about 15-20 minutes, finally rinse the bleach off with sterile water [pressure cook it]. Everything needs to be totally sterile, plants don't grow fast and any bacteria in there is going to take over quick. Are you adding sugar to your media? What kind of vitamins and nutrients? [you'll need at least some B vitamins] What gelling agent are you using? Do you have a clean box?

I was going to make agar with coconut milk and condensed water from willow trees. I will have to test the Ph of that.

I was going to Pressure cook the agar half a inch thick in
quart jars with tyvek filters.

The coconut milk will probably add sugars also I think.

All work will be done in a still air box.


I might start looking for a cheep source of premixes agar with the root and shoot hormone in it if this is just a disaster.

Just a heads up I haven't seen anyone have success with salvia leaf cuttings, which could make it difficult for your micro-propagation experiments.  I've got a friend who has a salvia leaf with a shitload of roots but no shoots and I've never seen anyone get shoots from just leaves.  If someone can prove me wrong about this, I'd be pleasantly surprised.

In any case maybe you know this already and just want to figure out how to get shoots to form or somesuch thing.  Good luck :smile:

I think getting premixed hormones/agar, or just pure hormones, etc. might end up being way less hassle if you are serious. I mean you have no way to know what vitamins the plants will/wont have, how much sugar, and levels of hormones. I've had success with Salvia nodal cuttings in MS media w/ vitamins, 1mg/mL PPM, 1mg/mL BAP, and 5% sucrose. I haven't been able to get shoots from just leaf cuttings either but the stuff grows so fast, and will put out roots without any hormones in culture it's not such a big deal.
