vacutainer question

I have some LC in vacutainers from which I aspirated as much of the nutrient water as I could and replaced it with distilled water (per some of agar's old posts regarding long term LC vacutainer storage). I was having a bit of trouble doing this as the needles of my syringes aren't long enough to reach the bottom of the tubes. How careful I need to be about aspirating all of the nutrient water out. If there is still some remaining when I add the distilled water will the mycelium still go into stasis (that is once it consumes the remaining sugar)? On a side note the LC had very little sugar content and made with distilled water. Is replacing the LC with dH20 even necessary?

I have also been having a problem with some of these tubes going green after about 3-4 months, I am assuming that I probably introduced some contams when I replaced the LC with dH20. Is there a chance it could be something else?

Agar slants, stored in a refrigerator is a much better method.

I'm beginning to realize that. How many slants do you keep to ensure nothing gets contaminated? I've been thinking for a while that I would like to move to store to them.

It's not a problem of contamination..once you have a pure culture and you've the skills to achive it you shouldn't have the problem to make slants.

You make more slants to be sure or quite sure to have multiple clones of that strain, in the case you lost one vial (for example it drop to the floor and crash and so on).

Remember to test the viability of the mycelium in the slants. Insert into the slants a pair of tootpicks or wood in general like RogerRabbit suggests.

Fridge at 4°C is good. Mines are at 8°-10°C because my mom uses that temp..the mycelium grows however a bit at 8°-10°C
