Agaricus silvaticus Questions

Hola I recieved a few edible prints from anno around a month ago and decided to try and cultivate some of the strains he sent. I made a batch of agar and poured the plated about 2 weeks ago and on the same day I poured them I innoculated them with Agaricus silvaticus and Enoki spores. All of my enoki plates eventually contaminated with bacteria, but the Agaricus plate is contam free, the only problem is that the spores will not germinate. It has been over two weeks now and I have no signs of anything on the plates. Any advice on getting them to germinate?

what agar mix are you using? Is it peroxidated?

No it has no peroxide. I made the agar for cubenis primarily, it is a mix of powdered straw, PDA and gubba juice instead of water (gubba juice is what I call the water left over when you pasturize horse manure). It was sterilized as usual. The formula workes great for Panaeolus and Cubenis but nothing else will germinate on it. I am going to be doing some more plates today with just PDA that I got from the sporeworks. The stuff I used before was home made with food grade agar and powdered potatos lol.

that's strange, i have no idea.
I use food grade agar, usually.
my recipe is
9g agar 5g powdered potato flakes  10cc's karo syrup 1.5g yeast 500ml water
then if i'm peroxidating it, i take out 4cc's of water
wait until it cools, and use 4ccs of peroxide in it.
It works well for me.
I'm going to be working on silvaticus soon, i'll let you know how my results turn out.

I also had some bacterial contamination, here?s a solution though: When you streak the spores on agar, make sure to only take a small amount of spores from the print and try to spread the spores over a big area of this dish. Eventually you will get clean growth on agar among the bacteria contaminated, and you can isolate it.
Or use antibiotics agar if available.
If you still have difficulties, I can send you a clean agar wedge.

I have almost used the whole print now. I have inooculated alot of plates, I think 7 total now with the print. I have no inoculation loop so I have been using the a syringe needle to do the inoculations (makes it hard to streak ). I ordered some individually wrapped and sterile inoculation loops from e-bay yesterday so maybe I will wait it out until those arrive. My local grocery store sells enoki mushrooms but I don't know what to expect from the strain selection or their packaging process. I just made a new batch of PDA with peroxide tonight so I may go and pick me up some mushrooms to clone tomorrow. Thanks for the input both of you, I will update my situation when it progresses a little.

Ghetto, yet effective. just how I like things I like the idea of using a new sterile innoculation loop every time. Helps reassure me that this is not going to be a problem contamination wise. For the time being though I may have to make one of those, I may even put a wooden handle on it if I end up liking it Thanks

It should be somewhat thinner than a paper clip and alot longer so that your hand is far away from the plate when you are streaking.
