lc clone?

hi i just wanted to make sure since insomnia affected brain can no longer calculate. if i put a mushroom stem in lc and innoc jars with it would that be same thing as single strain cloning? and how would i sterilize teh stem in a glove box? wipe it down with alchohol?

I recommend CC(creamcorns) method of putting together LC, one of the best changes from the norm. is instead of trying to carve some mycellium out of the stem you simply take a needle and stab the base of the stem (a biopsy) and then inject that small piece of shroom you now have in the tip of your needle into an LC.  As far as cleaning up the shroom IDK you'd have to ask someone thats used the method recently.  I usually pick mine wearing gloves and have everything ready and swab everything down with alcohol but I usually dont mess with the actual shroom itself very much.

as for cleaning the actual sample the tek does suggest swabbing it down with some rubbing alcohol. im kind of on the fence about this myself now as i've played with some other things, i might be revising that soon... alcohol damages the hell out of mushroom tissue and you get such a small sample via biopsy that it leads to less than perfect success rates

i have had success making a half/half bleach/water solution in a tall shot glass and simply dipping the stem in there for about a minute

the idea is that the needle takes the sample from inside the mushroom, but there is a small chance there's something nasty on the outside of the stem you pick up as you poke it, hence wanting to clean it somehow first

other ideas might be straight peroxide (though i got a feeling it wont work as well as the bleach water) and iodine... neither of which i've gotten around to testing enough times to come to any conclusion to suggest for sure... ymmv

if you have a glove box by all means flame yourself a scalpel or exacto knife, split a mushroom open, and cut out an interior tissue sample that way... obviously inner tissue is clean already and hasn't ever been exposed to the outside world. better success rates, but it does take away from some of the simplicity of the original tek

You can pour iodine over the stem to kill live bacteria on it, but it's really not necessary if you rip the stem lengthwise first to expose tissue that has never previously been exposed to air, thus generally free of contaminants. Outdoor mushrooms should be dipped in iodine no matter what before cloning, but with indoor grown ones usually it isn't necessary. Don't use alcohol on mushroom tissue.

haha looks like we were posting at the same moment - but there you have it RR gives the green light on the iodine so i take his word its a viable option thats less destructive than the alcohol :thumbup:

how about dip it in beach water then rince it with peroxide? i'm gonna try both methods of needling it and ripping the sample out. thx guys.
