Cloning, Agar work, and my pre-requisites...

l know how easy it is to clone pot from a mother plant, for a perpetual harvest (or to spread the bloodline around)...
when i first grew pot (or even looked into growing), i knew the investment in true-breed seeds was something you only want to do once. $50-$400 is alot for 10 little seeds (esp if not feminized) and that's wy cloning is the OVERLY OBVIOUS method for self-sustaining.
so what about for shrooms? i know about making spore prints & homemade syringes i know about LC i know about grain-2-grain transfer (which never sounded like my forte) and i know LOTS about buying retail syringes.
and i ask myself... "i KNOW that i'm only going to buy seeds ONCE to get a stable vigorous mother, but i continue to spend hundreds upon hundreds of dollars for vendor spore syringes. what is the equivalent in the mycological realm for cloning and perpetuating?"
if that made any sense, i would LOVE some feedback.

I dont get it..
Your question: "what is the equivalent in the mycological realm for cloning and perpetuating?"
The answer: "I know about making spore prints & homemade syringes i know about LC i know about grain-2-grain transfer (which never sounded like my forte)"
I mean you can clone in the mushroom world too, but taking prints is the main way of perpetuation..there is break down that occurs in mushrooms after a few generations of cloning, so you always resort back to printing for perpetuation of your grows.
You also think of it as keeping a "mother"..generation 1 from spores..from spores, once you get a nice fruit, from a strain that performs well for you under your conditions you clone it. Thats done by taking a peice of the mushroom tissue, any peice will do however you want to go for the stem, and the inside of the stem. Then put that peice to agar or grain or a LC, anything to make it grow out. Then you go from there and use G2G transfers and liquid cultures to knock up more and more jars. For instance, take a small peice of tissue from a mush and put it into a LC with 200ml..when that forms a nice cloud of LC, you now have your "mother". You can store it in a fridge and fill a syringe as needed, or fill 20 syringes and store them all, or maybe put 10ml into a vacutioner or something so you can go back too it. Either way, thats enough to knock up 200 jars, and each of those jars can be split into 10-50 (I usually do a dozen) jars by grain to grain transfer working out to 2000-10,000 jars of grain, inoculated with the same mushroom.
Spawn each of those to poo and your talking wayyyy more then a lifetime supply of mushrooms. Mycelium doesnt last forever, months, perhaps years, and you cant keep taking that beyond those generations in which I talked about. If you reach any one of those limits, you need to go back to spores and start over again. Its not as easy to keep it going for a decade or so like plants, but its easier to get a woderfully producing strain from multispore, let alone clones (it only takes one clone, not many generations ect, and a generatioin for mushrooms is quick).
Hope I answered your question..

so if you are going to clone a prime shroom to start stock from, you can take a tissue sample from inside the stem and put into an LC? does Karo work here, or is malt/dextrose a better choice?
what about with agar? you just take a tissue sample and knock the agar petri?
also, when you're going to take a tissue sample (in a glovebox i'd imagine) how do you get the shroom into the GB? can you just wrap it w/ saran wrap or put it into a sealed jar - then sanitize everything inside, then unwrap/open?
sorry if these are obvious questions, but i haven't branched out this far yet.

Yes, Yes and Yes. You can actually just put the shroom in the glovebox either way. Cut the mushroom open, take a sample from either the very center where the stem meets INSIDE the cap or at the base.
