break up the myc with your marble or whatever you used, insert the syringe and suck up the little strings into the syringe

an LC is ready to use when you wanna use it. :smile:

you can let it colonize more certainly.  at this point it should be growing at a pretty good rate so a few days will make a difference.  as long as you leave some mycelium behind when you draw your syringe, it will continue to grow (until nutes run out of course) and replace what you took.

when sucking it up yes its best to break them into smallest bits possible, but be careful when agitating an LC that you don't contaminate it.  depending on your lid setup, you don't want liquid touching your filter material.  my preferred way, is to stick the syringe in, and suck up liquid... then forcefully push down on the plunger so you squirt a jet of water into the cloud... repeat once or twice if necessary... that will break it up into tiny bits no problem without risking getting your filter wet... it will also disperse it all over the liquid, at that point i just slowly fill the syringe up trying to point the needle at the bits and capture as much as you can.  you can certainly shake the syringe up a bit more once its full too.
