can the disposeable polystyrine petridishes be steralised in a pressure cooker after PDA or MEA have been poured thanks reply quick time is an issue!!!!

no, they'll melt, also you want to pour after, it's a fucking mess and a bitch to clean *when* it gets everywhere

so should i steralize the agar in say a beaker for ohh 30 mins at 15 psi?

If you need to remelt your agar you can use the microwave. YMMV.

FDA says autoclave 15 min at 121°C for both PDA and MEA. That's for 1L, increase time for larger batches. They also say that PDA should not be remelted.

no no my agar is in powder form so remeltings no prob.. hm mycosupply said mix 40g to 1liter and then PC for 30 mins at 15 PSI

Well, you should go with the manufacturer's directions unless you're doing government work or want to publish your results as using FDA standard methods. I usually round up to 20 min just to be safe.

I use a liquor bottle to sterilize agar. You can stuff the top with polyfill, then cover with foil. Pour the dishes after the agar has cooled to the point you can handle the bottle without burning your hands, but before it has solidified. I use 40 minutes in the PC at 15lbs for a .750 ml bottle filled 3/4 full. That is the perfect amount of agar for a sleeve of 20 dishes.

Don't open the sleeve of dishes until you're in your glovebox or in front of a flowhood.
