I have access to a production embryology/stem cell research lab in the evenings for use of their flow hood and autoclaves, but I am very concerned about putting a heat-sealed bag of rye berries and water into a $8000.00 machine and having it explode on me... the machine or the bag? has anyone used a laboratory autoclaves with the common filter-patch bags (these are from fungi-perfecti) before? These units sterilizes at 30psi/240degrees (f) (I think) for 20 minutes? Are these bags up to this? Do I have to ensure that nothing blocks the filter patch to allow steam and gasses to escape the bag? During the sterilization process is pressure inside and outside the bag equalized such that no bag expansion due to gas/steam inside the bag will occur at all (i.e. my concerns are totally unfounded)?

My friend uses a pelton and crane autoclave with spawn bags (28psi for 1 hour). If you seal the bag, it will pop and you will have a nasty mess to clean up. My friend puts the bag with no more than four cups of rye in a bread pan along with the proper amount of water and gypsum. Fold the top of the bag back and fourth until it lays nice and flat. Take some cotton string (not nylon!) and tie a few loops around the bread pan to keep the bag in shape. Cover with aluminum foil and cook. She has not had any problems at all. After the bag has cooked, remove it, seal it, and shake it up while still hot to distribute the carmel crap at the bottom. Once the bag has cooled you can innoculate with a syringe.

My friend places two layers of tape, about 4cm square, one on top of the other, on the side of the bag before she autoclaves. When she innoculates, she lifts the top piece of tape and pierces the second piece of tape with the needle. Drop the top piece of tape back down after the needle has been removed to seal the hole. She uses two pieces of tape because it seems to help add a bit of structure to the injection site and makes it easier to pierce the bag.

If your autoclave is bigger than my friends, you can probably forget about the bread pan. My friend has to use it or the bag will expand and touch the sides of the autoclave causing the plastic to melt resulting in another mess.

Also, when the rye is cooking there isn't too much smell, but when you vent the autoclave it will smell like a bakery.

Wow! That is one of the most scary things I have seen in a long time. If I found out that my doctor was using a mirro pressure canner to sterilize their surgical instruments, I would friggin freak!

Man that is some pretty weird ass shit they sell on there...any place that sells rectal and vaginal speculums that are gaurded by a cat named Lucifer is pretty weird....thanks for the laughs though

No joke. Thanks for the data, guys. When I consider the fact that the grain 'cooks' a little during the sterilization, leaving the lab smelling like a bakery, I think I will buy one of the American 25x Electic stereoclaves for use at home... Just like Seuss said he would be pissed to learn his doctor was using a mirro pressure cooker to sterilize surgical instruments, the fertility clinic's patients (and some of the staff) would be less than tickled to find that I was baking 'rye bread' in their autoclaves in the evenings.
