problems with agar work

I am fucking up somewhere in the process of isolating the strongest mycelium from agar with a multispore inoculation.

Of three attempts, zero were succesfull. I seemed to be able to get the rhizos by themselves, but having trouble fruiting.
I've read isolating a non-fruiting mycelium is un-common, esecially 3 consecutive times.

I've no problems colonizing jars, spawning, fruiting, LC's, or anything else. Just fruiting from isolated mycelium?

Any indications of non-fruiting mycelium (if in fact this is my problem)?

Are you using the isolated mycelium to actually inoculate something or are you trying to fruit directly of of the plates? I'm a little confused based on what you said.

Innoculating liquid cultures with myc off agar

No prob when innoc lc from agarmyc when isolating myc from contamed print (wild etc) - only when i try an isolate rhizos/strongest.?

Not all isolates will be fruiting isolates. That's why multispore inoculation works to a certain degree most of the time. You'll want to isolate at least ten different rhizomorphic single sector isolates on agar, then fruit out each one. It's common for me with ten isolates to have three or four that don't fruit at all, three or four that are mediocre fruiters, and one or two that deliver prolific flushes. You have to hunt for that really great strain.

That's why I prefer to transfer all the clean growth from agar to the jars, and then clone the fastest/healthiest/biggest/best fruits of the first flush.
