rye grain for incubation from spores ?

Hi, (please excuse my plain english, I speak french first)
I just read the Paul Stamets book, The mushroom cultivator. In his book, he use rye grain for incubation but he start the mycelium from agar culture and then transfer the growing mycelium from petri dish to grain media. (rye grain/water) Just want to know if I can bypass the agar step and inject the spores directly from the seryngue in the grain mix. (rye grain and water paul stamets mix). I know that the pf tek directly inject the spore in the grain media, but it's not the same mix and I already own some rye grain! Thanks a lot

Do you have a pressure cooker of any sort?

Seed/grains must be soaked first (18 to 24 hours), rinsed, drained, added to jars 2/3rds full, filter type lid, then pressure cooked at least 60 minutes - to sterilize the content. Then (once cooled) you can inject directly with a spore syringe.


Above is a wild bird seed mixture, two 1/4 inch holes in the lids & tyvek under the lids (polyfil will work). Coffee filters are used as exterior dust cover. #5 is after the jar was pressure cooked.


6 days after injection (incubated 2 80F)

Agar,
Yes I have a good pressure cooker. I don't add any water in the grain once soaked, rinced and drained ? Only fill the jar to 2/3 and autoclave at 15 psi for 1 hour ? The paul stamets spawn formula is this one, for 500ml mason jar :

120ml rye grain
90ml of water

Mix both and presurize 1 hour at 15 psi. I have try it and this fill about 2/3 of the jar. I want to be sure how to proceed, and wich one of the spawn if the best. Thanks a lot

Stamets forumla should be fine but I would PC for 90mins to be sure. If you follow agars advice it allows for germination of some endospores and the moisture is in the grains already, then you can get away with a 60min sterilization. If you add water just before PCing you must allow time for the heating grain to absorb water.

My wheat grain absorbs very little water in 24hours so I would have to add more. I don't think agar adds extra water after soaking. Others simmer the grain which allows for more water to be absorbed.
