problems with autoclaveable bags
    #4368306 -

hey there!

i got problems with my autoclaveable bags...
somehow i got many bacterial infections...
lemme describe the way i work:

i take my rye and soak it for 24h in water (so that the endospores germinate).
after 24 hours i take em out of water and wash em with tap water...
then i slightly boil the rye for about 30 mins.. until i see the first corns bursting...
then i take em out again.. wash em.. and let the water drip off.. for about 30-45 mins...
after that i put the rye in my autoclave able bags.. pull the air out and they get sealed with an impulssealer...
then the bags are put the autoclave (as much as possible.. else the seal will burst)... and they get sterilised for about 2 hours (more than 2 hours) at 121?Celsius - 124?Celsius...

they get innoclulated with agar syringes (which are fine... i try every batch on a lil glass of sterilised rye.. colonizes perfect)

but somehow like 50% get bacterial infections... the rye gets slimy... and mycel wont grow..

someone got a advice for me?

thanks in advance!

I would bet the rye is to moist. Don't BOIL it. Just "steep" in hot water, after a soak. Then, allow to drain - VERY WELL.

You could also mix in 40% (or whatever) WBS to the rye. That gives you differing size & type seed/grain in the mix. Which is more forgiving as to moisture content. Some will be a bit moist, some will be a bit dry. It all balances out - better than rye alone.

hey agar!

thanks!
forgot to write that i use birdseed too!
yep.. i use 60% rye 40% birdseed...
but well.. seems like i have to let it drain off much longer!
well.. i dont really boild the rye (my english is just to bad)
i heat the water to about 60-70?c and let the rye in for 30 mins...
the birdseed i just soak for 24 hours.. and then i mix it with the rye

thanks again ;]
i hope you are my hero heheh

Do you know how much water you are adding? i.e. wiegh the dry grain and then wiegh it after PCing to see how much water you have added.

no i dont :x
im always makin rye/birdseed mix for 2 bags...
thats like...
2000g rye
1200g birdseed
i soak em separated
the rye gets a hot water bath
the birdseed not
both get drained.. and afterwards mixed..
then i spread it to 2 bags...

Might even just want to dry mixing 20% dry verm into it after draining, it soaks up any excess water very nicely.

i just got off the phone with fungi perfecti and i talked to this guy who I GUESS WORKS IN THE SPAWN LAB and he said they just add water before pcing directly into the bag and let it sit. i dont know if this helps but he suggested 2000 grams rye to 1700 ml water. they let the bags sit for 1 hour to sometimes 4 or five hours. then pc. thats what they do i guess it probably works.. i ll be trying this technique soon for about 100 lbs of agaricus blazeii spawn.
