Petri dishes and sterile procedure

I've had a considerable problem with condensation buildup on the inside of the petri dish lids. The water droplets fall to the agar and disrupt the mycelium growth. I've tried stacking the dishes in stacks 9-10 petri dishes high. This prevents condensation from building up, but only at first. A few days later, the water droplets start to form. This hasn't caused any contamination problems, however, it does reduce the amount of mycelial tissue that is availble for transfer to rye jars. I suppose I'll have to wipe off the lids with clean tissue paper, this will increase the contamination rates. Does anybody have any ideas? Thanks a lot for the help, I'm a newbie and can use all the help I can get!

you can use an alcohol wipe, do this in a glove box and let the lid dry before putting it back on.

I wipe mine sometimes before I take pictures, and sometimes before I use them (after cooling). It has not increased contamination rates one bit in my situation. As hawk said, wipe them in a sterile work environment with a sterile cloth. The dishes picking up condensation even when stacked indicates you have a large temperature fluctuation problem. You should try to get that under control, as it causes complications for colonization.

With temperature fluctuations you can get zonate growth on species that do not typically exhibit this characteristic. The more stable you can keep incubation temperatures the more constant growth will be, and you will have virtually no condensation...

What do you seal the petri's with?

I use a medical tape called micropore.

It is like hepa filter tape & allows some gas exchange, so condensation doesn;t occur.

Not sure if that would be a wise solution. Condensation could still occur, and you will have a big pool of it on the lid. Then when you turn it back upright to examine it, the water would run onto the culture. This could flood the culture or spread contamination...



I forgot to mention that the amount of agar you pour in the plates has alot to do with the amount of condensation. Try pouring them thin, mine are usually only about 3mm thick...
