Agar - Will a fully colonized plate be a problem?
    #340421 - 06/14/01 06:34 AM (15 years, 10 months ago)

Hi everyone,
One of my plates have been completely colonized with rhizomorphic mycelium. In TMC, it says that a plate should only be colonized 2/3 ( or 3/4 ) of the way. Otherwise, contams from the sides of the petri could inhabit the mycelium and go undected.

My question is:
Would cutting only the mycelium that is 3/4 of the way away from the edge work to only get clean agar? + Would it matter if I was using Peroxidated Agar ?

-the sweet is never as sweet without the sour

Cutting from the center will work fine. Also, peroxidated agar is no good for germination. You can use it w/ clones and isolates, but I wouldn't do it unless it's necessary.

Where there's skill, there's a better way..
