agar questions

i was wondering on how long agar lasts, and what conditions are necasary to allow it to last to its fullest potential. this goes for both colonized agar and uncolonized agar. thanks so mush (pun intended) for your time ;)

agar can and will last for a very long time if you take care of it,
if you are using petri dishes get some black stretchy electrical tape(3m makes the best stretchy tape) and run it around the edge sealing it to prevent it from dehydrating and getting contaminated. if you have mycelium on it then store it in a cool place below 60 degrees F

To tape the dishes and culture tubes we use (a dutch version of) Saran wrap. With a sharp knife small roles (2 cm) can be cut from the big role. This tape is self adhesive and allows gas exchange.
I got the idea from a plant tissue culture lab where i have worked. They used it instead of Parafilm because it allowed better gas exchange and was easier to work with.

It has to be pointed out, that the material is not the same as Saran wrap( which is polyvinylidene chloride (PVDC) and is NOT permeable to gases and water vapor) but polyethylene(PE) which IS permeable.

Does anyone know of any particular brand of cling wrap that is made of polyethylene(PE), cling wrap sure would beat the price of parafilm, right now I'm using 3M's Micropore tape.

ooops...my mistake. The wrap we use is indeed made of PE. Every supermarket here has it's own brand and they are all made from the same material. They all seem to work fine.
This stuff is lot easier to work with than parafilm. Parafilm has to be cut into right sized strips. While this tape can remain on the roll and does not need to be precut.

ziplock bags don't keep the dish and lid together. By using tape you can never accidently tip the lid off.
