silly agar questions - new trials beginning

hi everybody! lil question... how long do you let your mycelium on agar before transfering it to a 'virgin' plate? (in the initial transfers, of course)???
oh, and a PS: when you talk about yeast are you talking about cake baking yeast or beer brewing yeast? (or are they the same?)
PPS: is it bad if i get bubbles in my agar when solidified?

1) After fully colonized, you want to use the best mycelium
2)Brewers yeast is higher in nutrients, so go with the brewers
3)No, it just don't look as pretty

When to transfer really depends on the cleanliness of the germination plate, or the original clone plate. If it is clean, you can wait until clear sectors have developed. If it is dirty, transfer as soon as you see something clean.

yeast extract is dead yeast nutrients. i was under the impression you were supposed to use that.

Another tip that wasn't mentioned: Remember to slightly alter the composition or your agar formula when transfering a strain from one plate onto another. This will insure a healthy strain and allow the mycelium to evolve rather than degenerate.

nighted: what you mean by slightly alter the composition of agar formula? im using PDY, you mean i can alter this formula slightly or alternate to MEA?

Yeah I do that as well...can be as simple as adding a little BRF or honey, or whatever that has nutrients that mycelium love....cheers.

Actually, altering the substrate will slow the fungus down initially, since it has to "learn" how to digest the new substance. Instead, diversify your agar mixtures from the beginning:use PD, malt, and whatever substrate you intend to use for spawning( if you add dung extract, make sure you correct the ph and don't use too much as a high concentration is sometimes toxic for the fungus). This way, the agar wedge will not take long to recover when transferred from the agar plate to jars of substrate.
If you want to help prevent your strain from degenerating, make sure when you germinate the next generation from that batch that you change the agar medium. This is by no means a guaranteed way to stop mutations, but it seems to help from experience.

When you say switch it up, do you have to right from the start? say i put some tissue in there, grow it out, then want to transfer a section to a new plate....do i have to change the agar already? or does long term use of the same mix make it degenerate?
