gar.. dog food agar questions ( silly obstruction..)

Ok, i was planning on using rush wayne's peroxide method with dfa
but i have run into a very simple problem: i have no scale

does anyone know of an average density for dry agar agar?
ill get the volume of 20g dry dog food just by dividing total weight by volume and so forth.. but i dont know the weight of my agar at all

and also

using the peroxide method, how important is it to have sterile h2o2? Is there some way I could check the sterility? I have some mildly old peroxide, but is it ok to use?

You don't need sterile H2O2 -- after all, it's there to kill things. :wink:

So long as the peroxide was stored in a cool, dark place, it should be fine for a year or two.

aye, but i read that sometimes it has mold particulates floating around- sort of establish mold mycelium which would contam my plates

Not much will live in hydrogen peroxide. The volume of the agar depends on the specific agar, but you could always do it by the amount of water displaced.... You'd have to know the specific density, however, but this may be on the bottle.

A drop scale should work, or a nickel weighs 5 grams. It's not really sensitive, but you want to be close.

BTW -- dog food agar contaminates really easily, IME. I don't use peroxide, however.

interesting.. i've noticed some places that recommend it, and have used it successfully for morels and other interesting fellas.. im going to use it on pl. djamor ( most likely), so i shouldn't run into contam problems, esp with peroxide

Doubtful you'll have problems with peroxide. Like I said -- I don't use it, so I have to worry about what I use more.

I will use gentamycin sometimes, though.
