Hoping for some help to save my GT's soon from pinhead mold

Ok. I'm doing my first grow but I've done alot of reasearching before starting so I made sure I followed procedures and sterilization (small room with no carpet, 2 Hepa filters running for 2 hours before, grains soaked for 24 hours, simmered, then PCed, sterlize needles and all tools with alcohol and a butane flame, ect) and I havnt got a contam except this strain.

I started growing about 3 weeks ago with 4 syringes from Ralphs. Z strain was done with GT's and Z strain has gone on to colonize, are now cased and on day 3 of fruiting and grain to grain transfers to 8 new jars were done which are almost done. I've also done since then cambodians (first batch will be ready for transfers to new jars and casing in a few days) and brazlians which have also been cased and the casings are incubating now and transfers done and new batch of jars are almost ready to be cased. I also did some Tampanensis a few days ago with no sign on contam and showing signs of myc today.

Anyways my point is I've done quite a bit of work the last 3 weeks with no contams. However this GT syringe kept producing green. I did 4 quarts, all of which got green, Z's done same way at same time and did excellent. Later on I did 5 petri dishes of the GT and no myc growth, only green. Green started in 2-3 days on jars and dishes. I left everything to continue growing (spereated from everything else in a different rubbermaid in about a 71*F room) and I got a nice white chunk out of 1 of the jars that wasnt near green. Well I transfered that and it was growing great for about 4 days, growth was seen after day 1 and myc grew thick and wite on top 1/4 of jar. Then all of a sudden I see this pin head mold I've heard of. Definatly is that, its an obvious spot. By the way, I did 8 other grain to grain transfers right before that (the brazilians) and they all ended up fine.

Well now I have 6 jars PCing and a petri dish with some MEA waiting as well. In about 2 1/2 hours they should be ready to go. I'm hoping to give it a better shot with multiple jars this time instead of one.

What I wanted to ask is if the pin head is realated to the green stuff. I thought I was clear till this showed up. Do you think I'll be able to transfer these well? There is lots of marshmellowy myc on the top and about 2 kernals of grain on top covered in the pinheads. I will get peices away from that and try to get some from the interior part of the myc chunk and not whats right on top. Anything else I can do?

This stuff spread quick, there were 2 rye berries covered yesterday, today its forming on about 80% of the uncolonized substrate (about 3/4 a quart).

Well sorry for the long post hope someone will have some info..

I just was looking at a contamination page and I think the green mold I had was the Penicillium as it looks EXACTLY like one of the pics. In fact I am sure and it says black molds are related so maybe this was due to a not quite clean transfer...

Well transfers were done yesterday. Not sure about some of the jars but I got a nice peice for the plate. The myc may have gotten contamed spores cause its mixed in but we shall see...guess no one has any more info on this but thanks anyway.

Based on having clean jars from the other strains, it sounds like the problem lies with the GT syringe. What you're doing is good; trying to get clean mycelium from a potentially contaminated mix. The downside is that messing with the mix could spread the bad stuff around, maybe even into your other clean strains. If you're careful, it can be done and is quite rewarding.
The green stuff is likely either Aspergillus or Penicillium, the two can't be distinguished except by a microscope. The pin mold is likely Rhizopus, maybe Mucor. It sounds like there may have been a very low level of mixed mold contamination in the syringe. There are many ways this can happen, and shouldn't reflect on the vendor. However, given your experiments, it does sound like the syringe is the source.
It sounds like you're doing your research, I'm sure you'll get the results you're looking for. Just be really careful working around moldy bits. . .that stuff spreads like crazy.

Well by pics on the site it looks like a Penicillium..It didnt really turn white so much first and I dunno just didnt look like the Aspergillus but I'm sure it would look different in other pics and I'm sure your right..either way it wa green and doesnt produce mushrooms :smile:

The pin head mold looked like Rhizopus as well and no it shouldnt reflect the vendor as I got excellent service and 3 other excellent syringes from him and I still recomend him. He would probably give me a replacement but I'm new and want to isolate it myself because I plan on doing this for a while and will have to some day anyway. If it fails I will contact him.

I use extreamly sterile techniques and work in different rooms then my good strains, always kept seperate...I am being carful not to distroy them too and I think I should be OK. I keep my two HEPA filters running in the room where I work when I'm working and then in the house all the time so this place is very contam free.

Well thanks for your reply...new growth started on all jars today, nothing on the plate yet...I am confident that ONE of them will be clean this time. I'll update this thread just in case anyone is interested and reading it.

Agar started growing last night, all jars are showing growth but 1 had the pin heads last night. Other 5 still looking good.

I guess since the pin heads like the grains the sugar didnt take to them. The plates almost done which will then be used to innoculate some jars of grain.

Should I be okey from spores landing on wedges and not germinating on the agar then germinating in the grain? Should I do another transfer to another plate first? I'm not worried about getting the quicker mycelium I just want the strain going so if I should be free of contams now I just want to go to grain, what do you think?
