Having issues with grain jars
    #29702131 -

So i follow the PGT tech.
I put yellow corn in my pressure cooker. Add corn syrup (my liquid culture is made with some corn syrup too) put then lid on and bring up to 15psi. Leave there for 30 mins. When depressurized i strain in a collander, let it rest several hours. Proceed to put in grain jars sealed with aluminum foil to protect the filter. I place back in pressure cooker. Bring to 15psi for 90-120 mins. Let that rest a full day. Then i inoculaye with my liquid culture. Attatched is the results. Ive had good success before. And still am. But out of a batch of 6 jars, one has what you see. Just trying to pinpoint and correct my technique if thats the issue.

Were all the jars inoculated with the same LC? Prepped the same way? Pressure cooked at the same time? And some are ok, while this one is not?

If so, I'd look towards clean work or filter failure. Something that could be unique to this jar.

You may have introduced something during inoculation. If something shows up in your other jars I would considering the possibility your LC has become contaminated.

Mold should have died during the PC run even under suboptimal application so this is coming most likely from your inoculant, failing filters or bad technique.

Failing filters and or bad technique would explain why not all the jars are getting contaminated. Show us a photo of your filters. How soon after inoculation does the mold appear and let us know how you
inoculate, e.g. in SAB/ Laminar flow or open air.

What type of SAB or flow hood, what type of injection ports, flaming needle? Any information that might be related.

Also: You are not mentioning if you vent your PC before pressurizing to 15 PSI.

I do not vent my PC after its ran its process. I let it depressurize naturally.

SAB is just a home made unit.

I always use a fresh sterile needle. Flame it. Allow it to cool. Pull up LC, then inject into grain jars

Takes about a week for this to show.

All jars are pressure cooked at the same time. So therefore the same way. All inoculated at the same time therefore the same way.

Ill look into replace my filters.

1 additional question, when pressure cooking the grain jar for 90-120 mins is the lid supposed to be tightly sealed? Should i be removing the filters during this process?

Additionally i know visuals only go so far but the LC looks nice and healthy. White fluffy goodness.

Couple things:

Venting a PC refers to allowing the PC to blow steam out for 10-20 minutes before putting the weight on. This makes sure the entire volume is full of steam, which is required for proper heat transfer and sterilization. Not doing so might cause a bacteria problem, but I don't think it would cause mold.

Also, the only way to know if LC is good is by testing it on agar. A visual check isn't a good way to tell if it's healthy.

What are you using as a filter material?

How many jars out of each run fail?

I screw my jars lids on tight with the filters during the PC cycle.

Micropore tape. And then of course the filter the LC jar lids came with. Covered with aluminum foil during PC to protect filter.

This past run was 6 jars. 1 did this.

But it seems to be every time i get 1 or 2 jars like this. Didnt used to happen at all.

Micropore tape as filter should have 4 layers and be replaced after 3-5 runs through the PC.

Change it to a better option if you can, SFD's are the gold standard and fool proof, other options like polyfill do work too but you have to do it right to be reliable.

If that's the case then it sounds like technique to me.

... if not then it's the non venting of your PC.

Not venting a PC will never cause mold problems

Oh dang i didnt realize this. Ive just been running 1 later. Wonder why its juat now becoming a problem.  Ill switvh out my filters and start using more layers of micropore. And make sure to vent PC. Hopefully with all 3 changes we can get back to 0 contam.

I wouldn't be surprised to see it show up in your other jars, especially if that happened after only a week. You can potentially contaminate an LC every time you use it. It wont necessarily appear in every jar but it will likely appear repeatedly if the culture is contaminated and you keep using it.
