Help! Very slow PE growth on agar
    #29700998 -

Hello, I've been attempting to grow APEs from spores on agar dishes but they're very slow. I first inoculated 2 dishes with spore solution on the 9/12. When it had grown a bit and showed so signs of contam, I did 10 transfers to some clean petri dishes and some of those got contamed but 6 or 7 of them were good. But it's been 17 days and the dishes are still not fully colonized. What gives? The only thing i could think of is the room temp, which ranges from 68-70 F. Should I get a seedling heat mat? Or a space heater?

This is the agar recipe I used btw:
For a **500 mL batch** (makes aprox. 20 dishes)
- 10 g agar-agar
- 12 g light malt extract (LME)
- 2 g activated charcoal powder
- 500 mL water

9/30 and some on 9/23

Hey Ligma,

Personally, I don't like the patterns showing up near the plate walls or the others scattered around the centers.

IMO they're contaminated.

Can you take photos with light from underneath?
How do you prepare your plates?

Hugs

A couple of things could be going on. Inoculating an agar plate with a syringe could lead to higher contamination than using a spore print. I can't tell if they are contaminated with my eyes. Higher temps could possibly help. But I wouldn't get a seedling mat or a space heater due to the chance of too high a temp and/or drying out the agar.

I would suggest when making new plates to cut back on the LME. With a very nutritious agar the mycelium growth will be slower. If you cut back the LME the mycelium will colonize the plates faster and also easier to differentiate between clean growth and contamination.

They look good to me.  I’d do as veggie suggested, but I’d leave that fully colonized plate alone.

Make some plates and cut back on the nutrient source (LME).. Take tissue samples no larger than a grain of rice from the leading edges of the cultures before they reach the walls of the dishes.  68-70F is a very good temp range.
