Ya, I do think jars like that do cause anomolies. I had a pan cyan jar that I tested some time back that had a large blue mass in it that read quite potent. But I do not know if the potency was from a pin blob in the mycelium or the myceliated grain itself. I should have tried to separate some of the grain from the blob and tested them separate, but didn't think of that until too late.

Not intentionally. I usually do pull the jars and place them in the dehydrator in the late afternoon. But that is just coincidental due to my schedule and not planned or 100% consistent.

I like this idea for another test. I could modify the schedule to try this with a couple of jars, but I think a dedicated project would be better so we can get a more statistically significant sample size. Seems like an easy design. A few dozen jars, wait 12 months, add h2o to some and nute to others while keeping some control jars. Compare results after another 6 months? Something like that.

FWIW, I consumed some pseudo sclerotia from a Myceliated grain (semp) jar that had been consolidating for 4.5 months. Dehydrated 2.5 grams and it was quite a potent trip. First time trying something other than cubes so I can’t compare apples to apples but it was impressive. Will have to try just the grain to see if there’s a noticeable difference.

So are we thinking that alkaloids by weight is actually increasing over time? In my opinion, it doesn't appear like it is. Or if it is, it happens within those first few weeks after consolidation.
You said regarding your first round of testing "though 7 days from full colonization is actually pretty much the same thing as 15 days from inoculation, so there is that" — so you had full consolidation at around 15 days post inoculation. And concentration appeared to peak 15 days after that (30 days total since inoculation), after which it appears to remain level.
Current testing appears to reflect this also.

Thoughts? Conclusions?

Sources... because this whole thread has shown that Yes the total MG /g of alkaloids is proportional to the amount of time

Oops, I misread what ABE means. What I meant to say was that the total alkaloids appear level. The concentration increases, but this is due to it drying out.

So, if concentration doesn't matter, which it does when it comes to extractions, then total actives peak soon after consolidation?

Yep. If total production of actives is the goal, 4 weeks or so seems to be the peak. On a related note if you were to fruit it, the total actives of the fruit is in the same ballpark.

Now the good reverend's pre-colorimeter experiments indicated a rather significant difference in total actives between different grains. A common assumption is that the grain doesn't matter much for potency. But those results indicated that it can be a factor of at least 3 between grain types. If the total peak actives of the mycelia to the fruit is anywhere as closely related like ReverendMyc's small sample of results indicate, then...

may i ask what your grain prep method for mycelated grain is?
i just harvested a batch of mycelated wheat. after 5 months it turned from 1000g start dry weight into 882g of final dry weight. 5g of the grain feel similar to 0,33g strong cubensis potency wise. i wonder why they turned out so weak compared to the numbers i found so far. my guess is hydration of the grain, because i had this happen with another semp strain before.

Naw, I can't really recommend quinoa. By the time it is hydrated enough to sterilize properly it is really difficult to work with in jars. It might be better in bags, but that is not my area.

I like it for being easy for the myc to ingest and gluten free, but in addition to the difficulty of work, some of my testers object to the taste for myceliated grains. It adds a slightly bitter taste. My wife and son think it is quite bitter even though it is barely noticeable to me. But once you concentrate it in a tea for gummies it does affect the flavor. So I am still seeking the perfect grain.

Also, to answer your question more directly, it seems to be more about the hydration than the time in PC. But it is all a precarious balancing act that just doesn't seem to be worth the effort, imho.

have you tried sorgham?  it's very easy to work with and doesn't get gummy/stick together.  gluten free and $1.15/lb here for organic in the bulk bins.

A little bit. I found red here but the feed store only knows it as milo. I liked it quite a bit. Just need to work with it a little more.

I will have to check the bulk grocery. They had organic wheat that I liked for its cleanliness, but not gluten free and really sticky when rendering it down.

the jars turned out alright, still a few lumps but all in all a good texture.
i think i can imagine what the subtrops feel when eating the quinoa. its really easy to digest and gives me a good feeling, way nicer than most grains.

there is still one question i have:
what do you do about the starch when making grain tea for gummies? does it turn the tea to pudding too for you when you concentrate it? i had that with wheat.
my idea to circumvent this would be extracting the actives below 70°C to prevent the starch granules from bursting.
what do you think about that?

That was definitely my problem with wheat by itself as a myceliated grain. Pure pudding extracts.

I have mentioned somewhere that I have been playing around with vacuum extraction at room temperature. That has been showing some promise. I have a bunch of labs to run this week that keep getting delayed by life. One is a vacuum extraction that I have high hopes for. If it works out, I will certainly be sharing and updating here.

The other thing is filtering. I have tried so many things and was pretty pleased with the latest trial. I used these and got what seemed to be a better result. Again, if my lab results confirm my suspicions I will share what I have learned about that as well. I may still add one more filtration step on the next run to see if I can get a little clearer tea.

The pudding problem seems to be the final frontier of conveniently processing mycelated grains...
i think it could be worth it to look into beer and vodka making, maybe malt enzymes are the answer to the pudding :cheers:

*edit: indeed alpha amylase will break down the starch and gluco amylase will break down the dextrins from step 1 further down to simple sugars!  that means no more pudding and no more glucose sirup for gummies as we make our own in situ :rockon:

one can buy this stuff on amazon in liquid form and just add a few drops after cooking out the goodies and filtering. there are also kits for making oat milk that will do the job.
