I've never used condiment cups myself, but I know DV8DUG have used them forever and his agar game is as solid as can be. Besides it must be at least as sterile as cling wrap and people use that all the time.

Cling wrap goes outside the plate tho. If it works consistently for some then I can't knock it. But it seems crazy to me.

It's takes heat to manufacture them and I'm sure it's made under pretty clean conditions, then comes the hot agar mix. I have never had a poured, unused plate get contaminated.

Then there's the smashed wedge where a piece of colonized agar is sent in a zipbag. I have received plenty and only once have I had a problem, which could be my own fault, point being it seems plastic is pretty sterile.

I do agree with you though and is why I use disposable plates and wrap with parafilm :rockon:

Ziplocs are def sterile inside and I've used them in lieu of wrapping plates. I can dig it. Though I also kinda dislike smashed agar on general principles. That's just more boomer talk.

:lol: Boomer or not I'm right there with you. I like that with a wedge you can get a great culture straight out the bag, but I would take a dried grain over smashed wedge any day :awesomenod:

@Thought Criminal, we haven't seen you much since you started this thread. I just wanted to say don't get discouraged and don't give up.

Getting contamination early on is disappointing and a setback, but a minor setback that can easily be corrected by tightening up your sterile technique and sterilization habits. Besides reading and advice, the best way to get on track is from practice and developing good habits. You will get the hang of it in no time, I'm sure.

Keep in mind all of us have had contamination. It is normal and to be expected. Also not a big deal.

Also you can keep the LC on the back burner, at least until you get your agar game down pat. Quite honestly you can live without doing any LC at all.

Get back on the horse and practice. You'll be a pro before you know it. :thumbup:

I ‘nocked the LC with a LC syringe that I bought from a very reliable source. (One of the recommended ones on here) I know using agar is better but it was an experiment.

I used the honey recipe from on here, PC’d the solution for 15 minutes, used iso on the syringe and injected (though the SHIP on the jar lids) inside my SAB, which had also been cleaned with iso.

I honestly can’t think of where the point of failure might have been, so if anyone’s got a suggestion….  Regardless, I’ll be using agar going forward so I guess it doesn’t really matter anyway.

Not discouraged at all!
Maybe slightly annoyed at the turn out, since I thought I used good sterile techniques, but like you said that’s the way it goes sometimes.

I’ve been having good success with G2G transfers that I’ve been S2B that’s kept my fruiting chamber full all summer, so that’s been fun!

Mixed the agar, poured it in a glass bottle with a lid with a vent, PC’d it, poured the slightly cooled mixture into the cups in my SAB, put the lids on.

Only thing I can think of is that I didn’t clean the condiment cups. They were brand new and fresh out of a sealed bag, so I thought they would be OK. I’m gonna try again tomorrow and spray/clean all the cups with iso.

Any other suggestions I’m all ears. 👂

Don't do this. You will be promoting more contamination just buy separating them and rubbing them with anything including ISO.

How are you manipulating and using the cups from the sleeve up to your pour and lidding?

In a nutshell here's what I did when I used a SAB with condiment cups.

Wipe the outside of cups sleeve with ISO inside SAB.
Open the sleeve of cups and place a stack upside down on one side or the other of SAB.
Same with lids as far as wiping down, but place the opposite side of SAB and not upside down.

Pick up 3-4 cups one hand and 3-4 lids with the other, depending on space. Starting away from you place a right side up cup and immediately place a lid from the other hand on top. Set on top loose. Place all the cups in a row. Try approaching cups from the side only.
One at a time carefully lift the lid, pour agar then seal and 'click' lid down.
Done. Lather, rinse, repeat.

Limited exposure to outside influences. Gets around 95% success with practice. Remember to slow it down inside your SAB. It doesn't make you invulnerable.

Also, when it comes to actually using these, do they need some kind of air exchange? Like punching a small hole in the lid and covering it with micropore tape?

No. The air exchange is built into lid lock. No wrapping. When I open them up when they are in use, I do it upside down and with the lid cracked open. It's pretty safe that you can pick up a cup and keep it upside down if you want to work that way. So it's the cup sitting on the lid basically.

I do store mine vertically though...
