No pour agar water pooling advise? First time using agar!
    #29620387 -

Hello,

I've been searching through posts but can't seem to find the answer. Whenever I think I've found a post with the answer the post talks about something else and I started getting a bit stressed haha.

I've done the no pour agar thing for the first time. Actually this is my first time ever attempting to work with agar.

I used agar agar and light malt extract with water in screw lid tubs. Pressure cooked for 45 minutes at 15psi wrapped in foil with the lids slightly unscrewed.

I took them out of the pressure cooker and left them to cool and I've woken up to small puddles of water on the surface of the agar.

I've turned the tubs upside down for now.

I have a still air box.

What is the current advise about what to do with the pooled water?

Do I open them in the SAB just before inoculation and pour off the water?

Should I wait a few days before inoculation to check there's no contamination ?

This is my first time using agar so any and all help would be greatly appreciated.

I haven't grown for 5 years so I'm a bit out of practice. I've so far inoculated 10 grow bags using liquid culture I made from just a spore syringe directly into honey water and strangely there's zero contamination! Some honey water others are golden syrup and others are malt extract and water. However I want to start making sure I don't have contamination and having fun making isolates.

Best thing to do is lightly heat up the agar container by letting them float  in container of microwave heated water. Then as they slightly melted state, remove and stacked in your still air box. A few water drops doesn't ruin your culture work that much.

So put the plates into microwaved water. How hot are we talking? Couldn't I use kettle water as in boiling hot? The agar will then melt are then stack and it will reabsorb? Thanks for your reply but could you please be more specific about what I'm doing and what to expect.

Thanks!

Just leave them unwrapped for a few days in your sab.

I store mine upside down until I need them. 99% of the water will be gone after 2 weeks. If I need it sooner I just open it a bit sideways and pour out any excess. Just be mindful of good SAB technique and it will be fine.

So if I pour out the water that doesn't absorb but there's still a few drops left will it actually affect anything?

Just pour it out when you go to use them.
