To each their own as long as it follows sterile procedure and works.

heat sterilize whatever tool you plan to use, I prefer a wire loop.  Cool your loop by dipping it into the agar on your new clean plate.  Scrape the loop on the print, then open the plate and streak it in a zig-zag pattern.

In my countless attempts I've also tried that, but I don't like the idea of leaving pieces of agar on the print, I'd rather wait some time to make it cool it off.

But yeah, there's for sure room for improvement in my initial inoculation phase, especially about streaking.

Do you use a still air box?  What is your work space set up like?

Yep, I use a SAB, not much else to say. I lay a layer of 70% isopropyl soaked paper towels on the button. Unfortunately I don't have a dedicated room, so I'm using my kitchen, but as I said, so far, almost always contamination comes from the source.

I’ve been using sterile swabs to grab spores from prints. My caveman fingers seem to prefer that method. Lol.

So you basically just unwrap the sterile swab inside the SAB, use it on the print, then on the agar?

Are we talking about common swabs like covid test swabs?

How do you go about streaking in this way?

I think the cooling of the loop in the agar helps the spores adhere to the loop. As far as it staying on the print, I doubt that’s a big deal. People have been using that method for so long with great success so I wouldn’t worry about it.

Yep, pretty much. As far as streaking, I just lightly roll the swab on one section of the print, and move it in an S-pattern on the plate while rolling the tip of the swab.

My problem with loops has always been it kind of slicing the agar, as I always seem to go in at too hard of an angle.

Can you show us your still air box and setup?

Also DO NOT put a bunch of alcohol soaked towels in the SAB.  That's how you lose some eyebrows.  Alcohol fumes are very VERY flammable.

SAB work is based on the principle of still air, not sterility.  How it works is the still air in the box prevents particles from drifting into your work.  This is why it's so important to set up the box and then wait 30 or so minutes before starting work.  The movements you make in the box need to be steady, smooth, and with purpose.  Going slower is better.

The reason for asking to see your setup is so we can critique it and help make sure you are properly set up.  Your equipment or method (or both) are likely the source of your contamination and not the print itself.  No amount of of chat GPT can look at your setup and help you fix your methods.

I always use sterile swabs to inoculate plates with spore prints and LCs. You can use standard cotton buds. I cut one bud end off and fold the other end in foil and PC them in batches in a jar. I find the pressure you can put on a swab tip gives the spores better contact with the agar surface and if you use the same swab and zig zag a series of plates the spore load diminishes with each one and it's easier to get monokaryons off the last couple of plates.

I'm not aware of a specific writeup. I'm pretty sure I got the idea after reading a coment on here. It's pretty easy.

I use generic brand cotton swabs. I make sure the stick part is made of paper or wood, not plastic.

I cut one end off. You don't have to, but I like having one side different than the other so I can easily tell which side I have. This matters more if you're making spore swabs for long-term storage or to give to others.

I wrap each swab individually in aluminum foil. You don't need much, just small strips. The best way is to fold it in half, place the cotten end in the fold, and then just wrap the rest of it up.

I PC them at the same time as my agar. 30ish mins @ 15 psi. I put them in a jar when I'm pressure cooking them sometimes it has a lid sometimes it doesn't it doesn't really matter. I've used some that are months old and they're still sterile. I usually just store mine in a Ziploc bag because I don't want to have a clunky jar sitting around

I highly doubt you did, and why would that be useful to a newbie trying to establish basic skills?

Seriously, stop spewing chat gpt all over everything and just show your work.

EDIT:

Here, I'll go first.  Here's some standard cubes on a plate:

Here's some cubes on grain:

Here's some standard cubes fruiting:

I now have infinitely more credibility than you because instead of posting a chat gpt hallucination I ACTUALLY DID THE WORK AND POSTED IT.

OP, this is how you tell who to get advice from.  Dave has never posted a single picture of his cube work, let alone his process or repeated results over the years.  All he has is a sea of words that amount to absolutely nothing.

Trust who you want to, but I typically go with people who can prove their work.

Oh, while we're at it here's my SAB setup.

Super simple, dry, no alcohol inside the box.  Nice large arm holes so I don't bump the sides and my arms don't pump air like bellows as I move.

Yep, I know, but paper towels can be (and are) dirty. I put plates caps there during operations, caps that will then be moved over sterile agar, hence I think it can't hurt to have it soaked with isopropyl.

Of course, there are no flames inside my SAB...

OK so tomorrow I will try to both transfer the most rhizomorphic plates I got from my latest batch (the ones that went over the trench), and inoculate some new ones using swabs this time. Will keep you posted... and thanks! <3

That was not a stab at you, that was me saying Dave is a very bad source of information.

Anyway, looking forward to seeing those transfers.  And again, it would really help if we could see your SAB setup.  Kitchen counter is a perfectly fine place to do work, it's where I started.

Just caught this, this is where contamination is coming from.  DO NOT set the lid face down on anything, and alcohol is not sterile.  Alcohol can in fact introduce contamination to your work.  You should only be lifting the lid off the plate momentarily to inoculate or transfer, do not set it down.  If you do have to set it down, absolutely never place it face down on anything.

 Nothing fancy is exactly how I would describe a good setup.  I've fruited in Ikea containers before, if the shoe fits.

Nice advice! Will change that in my protocol, but honestly I don't think this is the cause of my issues. Otherwise I'd have come up (over the years) with a bunch of contamination randomly placed in my plates, and not only tied to the actual source / transferred agar wedge, which is basically never the case.

Just keep in mind poor technique works until it doesn't.  We had a mod on here do open air work to prove that very bad ideas can sometimes still work, but that does not mean it was a good idea or proper technique.
