So I decided to try some transfers, all of them using the trench method (first attempt). I basically transferred from 3 of the previous plates:

Follows pictures of the used donor plates.

From one of the first row, I tried to transfer just the halo:

From one of the second row I tried to transfer just a wedge of the border:

From one of the third row I tried to transfer just some ropes of mycelium using a pair of tweezers:

Notice how in the meantime, the halo started looking less organized, as if it started to turn into the tomentose white mycelium on top...

I also kept one of the plates as is, just to see what happens, and how the halo will evolve.

Sorry for veering the conversation away from Dave /s but here are some updates after 6 days since the above transfers.

Here's one of the plates from the most rhizomorphic one (third picture above) from which I transferred just one mycelium rope at a time using a pair of tweezers, you can see the mycelium successfully move past the trench. This is my first time doing the trench, now while I think this is pretty OK, in hindsight I could've dug a longer trench, comments are most welcome. Anyway I'll transfer again in the next days.

Transfers from other donor plates only displays some random tomentose mycelium, nothing worth to report at this stage. The only thing to notice is that the halo seems to have now turned into that tomentose mycelium. And this is confirmed by the plate that I decided to keep after the transfer (IIRC top right plate in the first image above).

As you can see the halo really turned into some tomentose mycelium, hence I'd say that the halo was just an intermediate phase at this time, for whatever reason. About it being cubensis mycelium or some other mold I'm not sure, even though I'd lean towards the former... what do you think?

Thank you for veering the conversation away from me! I think the halo was cube mycelium and it looks like it's making much better growth on the new plate. I'd take the next transfer from the sector at 3 o'clock in your second pic as it looks slightly more uniform than the rest of the plate.

Dave, see one of my comments in my air fryer thread when you have time. it's on the last page.

after all the discussion we had, i did some more research about heat vs no heat dehydration, and i've started thinking that no heat dehydration might actually be better.

i posted some of my reasoning in that comment, very briefly.

i am also interested in your reasoning for not cutting mushrooms in half during drying, as you've mentioned in a few comments. maybe i misunderstood part of your argument, so i'd appreciate it if you could explain it in a bit more detail.

I've never trenched before but that lil bit crossing onto the other side looks pretty decent, I'd feel hopeful about it.

This happened to a bunch of mine too, about 50/50. Some of them are clearly wispy contam and were never myc, but the ones that fleshed out look half decent, so IDK what to think either.

I got my congo red and a blue dye and a phone mount for the microscope in the day before shit completely exploded at work. Feel bad I couldn't get to it in time. But I definitely plan to return to those 12+ plates and see if there's anything conclusive in the scope.

Thanks for the suggestion, this plate is just a canary to see how it evolved halo-wise, I don't plan to transfer again from this plate, I'll perform my next transfers from those with the trench.

This is a donor plate from which I transferred, that exhibited tomentose white stuff in the center and the halo outside. I took a wedge out of the halo part, and kept the plate above just to see the behavior. So no, I don't plan to transfer from this.

I don't know if this is actually healthy cubensis mycelium or not, two things I'm pretty sure of about the halo in the OP:

  • it's not just bacterial contamination;
  • when it grows it becomes this fluffy thing.

I'd be interested in your thought process tho, what in this plate made you think it's some kind of contamination? Even if it's obvious, but I think it can be useful to me or others who read this thread. I see tomentose and uneven mycelium, with some yellowish areas in the middle.

Were you able to discern the wispy contamination by looking at the plates, or that's because they never got to fruit anything?

I've got a shitload of plates related to this project, so it's hard to keep track of individual ones. But when I was unsure earlier, I put a bunch of them under the microscope and marked the ones that had that reaching, more angular, slightly thicker seeming mycelium. Many of the marked plates that had distinct halos and were questionable early on have now fluffed up well, and become more orderly, to where they seem much more like tomentose cube myc than either of the mold presentations I've been running into lately.

It's still possible that it's two organisms on top of each other, I don't want to say anything conclusively until I've gone back and gotten a proper close look. I've kept all the plates, including the ones I would otherwise throw away, for comparison when I'm finally able to get to it.

I'd love to eventually see some side-to-side comparison.

:batlol::justno:

You mean the recessed part of the edge where it looks like it might have run up against a contam?

Take this guyโ€™s (chat gpt) advice at your own risk.

Personally I would toss out those plates if they are cubes.  They look pretty fucky.  Thereโ€™s nothing wrong with going back to spore and starting over.

it looks like it

disorganized myc. the yellowing? cubes grow like weeds and healthy cultures are easy to come by. they grow like crazy. so anything that looks off? is off.

i look for anything that is symmetrical. thats what i look to when i transfer from questionable areas. does the new growth grow out evenly? if not?? i either take another cut to see if i can get something more uniform or i start over.

its like a relationship. would you rather spend tons of time and energy on something that may not work? or just start over with something fresh?

sometimes tossing out the old can be the speediest way to gaining the new. cant fix em all kinda logic.

Thanks for this reply!

While I get what you're saying I doubt that starting over will fix my issues in this case since plates were like this since the get go (I mean, not exactly like this, but you can follow the progress in this thread). This means that contamination, if any, came from the spore print and it was impossible (or I was unable) to separate from the actual mycelium.

I think my overall SAB procedure grown to be fairly OK, I never see weird stuff except from the actual transfer, no random mold, no bacterial spots around the plate, etc. so this is getting super frustrating. These plates are the first batch of a new (bought) spore print, but in all my cultivation history I never managed to get perfectly organized and rizomorphic mycelium like the ones I often see here. Hell, if I had thrown away anything that wasn't like that, I wouldn't have grown any mushroom at all!

just hunt for organized and symmetrical growth and avoid chasing the rizo dragon. its just morphology or whatever it doesnt determine anything about the mushrooms that grow from the culture.

I've given up on Spore prints before too. Sometimes there's just too much contamination or what is there is just too aggressive

Eh, I'd love to... spore prints are currently my only source. I plan at least to obtain one healthy fruit, then clone it, and resume from there. Hope it makes sense.

Sorry for being imprecise. What I meant is I would give up on individual prints because they were too dirty.

One thing that seemed to make a difference for me is I started using sterile Q-tips to inoculate agar they seem to work better for me than inoculation Loops or anything else.

Your account is old enough for the marketplace if I were you I would just start looking there and try to gather up a few prints until you get one that's easier to work with. Just my opinion though. I totally understand the frustrations of just wanting to get something. I've been there myself

Do you have some reading material at hand? While I don't think it's rocket science, since we're talking about this, maybe I could avoid bumping in some outdated / wrong info.

FWIW I just hold the print above the plate, scratch it with a sterile scalpel, then I try to do some proper streaking, that's it. I don't know if this can be improved in regard of the issues I'm facing in this thread.

That's a nice idea actually, I always thought that the marketplace worked best in US and some other regions, not mine. Might be wrong tho...

Okay, do not hold the print over the plate.  That is dropping all kinds of contamination on the plate.  You don't want anything hovering over the plate at all if you can avoid it.

heat sterilize whatever tool you plan to use, I prefer a wire loop.  Cool your loop by dipping it into the agar on your new clean plate.  Scrape the loop on the print, then open the plate and streak it in a zig-zag pattern.

Only take the plate lid off when you absolutely need to, do not just leave it open for all of the work.

I'll dig around and see if I can find some of the videos that show this process.

Do you use a still air box?  What is your work space set up like?

Iโ€™ve been using sterile swabs to grab spores from prints. My caveman fingers seem to prefer that method. Lol.
