I guess what I'm saying is that if it's a bacterial Halo the Q-tip should pick some of that up and transfer it. If it's only digestive enzymes nothing should transfer or maybe a single cell or two will grow. If it's microscopic running mycelium then the Q-tip will hopefully grab some of that and transfer it.

I asked one of the people who was putting forward the idea that "embedded bacteria" cause halos to try taking a swab from the halo and grow it on a plate when the original scare happened. They did and there was absolutely nothing growing on the plate under the scope when they checked.

The thread is titled "Would love to hear opinions about this weird grow pattern in transfers." Opinions require explanations and evidence to back them up and that's what I've provided. And it wasn't that complicated or difficult to understand.

Some people are happy to hone their cultivation skills and master Teks and have no interest in how the Teks or the mycelium works and that's perfectly OK. Others want to know everything they can find out about mycology, especially as it relates to cultivation and improve their knowledge and that's OK too. One isn't better than the other because it's all down to personal interest.

DV8DUG, what have I said that makes you think I don't know shit about genetics? What have I've stated that you think is incorrect?

...and some of the genes that are expressed in reproductive mycelium are thought to stay "switched on" when the transfer grows out again on agar. So it's a well observed phenomenon that a clone culture from a fruit will fruit quicker than it's parent spore derived mycelium, for example, because a lot of the reproductive genes from the hyphae in the fruit stay switched on.

"In comparison to vegetative cultures, the excised fruit body tissues form fruit bodies very rapidly. For example, in Coprinopsis cinerea, renewed fruiting occurred within four days, compared with cultures inoculated with vegetative dikaryon which, under the same conditions, formed fruit bodies in 10 to 14 days."

I'm talking about your 'reproductive genes' and 'on/off' switches.

Yes, that's what he's talking about. And again here.

"Field-collected fruit body tissues of a mushroom usually generate abundant vegetative hyphae when inoculated onto nutrient agar plates. Such reversion from the fruiting stage into vegetative stage is not an abrupt process, rather there appears to be some sort of ‘memory’ of the differentiated state.

Initial hyphal outgrowth from gill lamellae usually mimics the densely packed branching and intertwined hyphal pattern of the gill tissues at first, being quite unlike the pattern of normal vegetative hyphae in culture.

The ‘memory’ could be no more than the residual expression of differentiation-specific genes before their products are diluted out by continued vegetative growth but as we have shown above there is considerable scope for epigenetic marks in fungi."

That does nothing to address this argument. He isn't even talking about Psilocybe Cubensis.

OK I think I got what you're saying, but I'm pretty sure this is no bacterial contamination per se at this point. AFAIK bacterial contamination always creates some kind of ooze, see pictures in this post, you can clearly see some kind of mycelium.

Alright, trying to wrap my head about everything that has been said here and what I wrote in my journal.

Facts:

  • I initially inoculated on water agar (and admittedly my streaking procedure was lacking);
  • since the germination stage I immediately started noticing those thin filaments and very few whiter spots, the trend looked like the plates in the OP: white mycelium in the center of the spots, thin filaments on the growth border;
  • I have always transferred from the growth border and very small wedges;
  • really fluffier white mycelium (again see photos) started appearing only on LMEA transfers;

With these in mind, here's what I think, correct me if I'm reaching the wrong conclusions:

  • the halo is not bacterial contamination because it's made of filaments;
  • the halo could be some kind of fungi that coexists with the good mycelium;
  • the halo could be part of the good mycelium that for some reason has that weird appearance.

I'm pretty sure I transferred micro wedges that (apparently) contained only some thin filaments fragments that eventually spawn the fluffy white one, hence I kind of lean toward the latter point.

In any case I'll do my best in getting rid of that before attempting to fruit. Will keep you posted!

I got daves first ignore i just cant with that guy anymore.

Bud, honestly your plates pretty much look fine.  Any grower with their salt will tell you that it's obviously good myc.  My only advice is to get into some regular plates so you can see what's really going on but frankly bud?

I'd fucking send those!

It has been my experience that changes in temperature can cause the halos in a culture. Faster/slower rate of growth as temp swings. Halo's do not immediately spell contamination.

My experience is that it does all kinds of weird shit and you don't exactly need an explanation for every single little thing. Tried and true processes combined with experience is what keeps the whole world medicated...

Wait what do you mean with "regular plates"?

While I tend to agree on this, this is the first time I see stuff like that, I mean it's not just some randomly uneven mycelium, look at that!

Anyway, thank you so much for sharing your thoughts. <3

Yeah, for sure, but IME it could explain ring formations and such, here the pattern is way too uneven IMHO.

I mean the classic recipe is 2% both agar and light malt extract.  I think normal petri dishes are always best for beginners.

Coprinopsis cinerea is a model organism in fungal research and many biological functions and behaviours are conserved right across the whole fungal kingdom even down to yeasts.

The observation of earlier fruiting from cloned material has been discussed a lot in various groups and it worked when I tried it with cubes. And it's widely known that you can invigorate a tired isolated culture by cloning a fruit. Try it yourself. It's easy to set up a side by side experiment.

And the pics of the angel wing clone cultures I posted above shows clear as day that the mycelium grows out in several different growth patterns from the same sample of fruit. That's because different genes are being expressed depending on the function of the hyphae within the fruit.

And you can see it here. The spawn cakes either side were taken from the top and bottom of the stipe and the one in the middle from the cap near the gills of the same fruit. But as you can see the cap culture in the middle is still producing psilocybin and the stipe cultures aren't.

Could you have isolated a monokaryon like Rasmodius? That would certainly explain the thin disorganised growth.

I think at this point you have some science ahead of you.

You need to transfer the halo to a plate and grow that shit out. Then we will all know...

This is my understanding as well.

Some pics of the plate that's like this, with what's definitely cube myc (unknown yet if di or mono) atop this other thing (which per above might also be cube myc).

Here it is intersecting with bacteria. The halo is interrupted and seems to be on the same ground plane as the bacteria, while the other cube myc is growing overtop of the bacteria. IDK if that means anything.

Re sending it, trenchhing, swabbing, etc, going to do all of that.
