This vacuum extraction looks really interesting. I'm about to do the Stoned Gummy tek, so decocted 3x with 100g of dry fruits with 2L water, man, reducing almost 6L down to 180ml took hours. One go under vacuum with less water would be great.

Thank you for the thoughtful discussion. It is so easy to misread malice in text where none exists. I think that we are closer to agreeing than it might seem on the surface.

I agree that what limited published studies are available seem to take the low heat drying as gospel. What I don't see in any of those studies quoted is a direct comparison of mushroom bodies dried at various temperatures and then tested for potency. They often discuss aqueous solutions of extracted psilocin and psilocybin. They clearly show that heat above boiling temperatures cause dramatic decreases in potency, but very little difference between room temperature and dehydrator temperatures.

I don't think anyone is arguing that psilocin is largely lost in the dehydration process. Even if it were not lost while drying, it is so unstable that it does not store well in any method accessible to most hobby growers.

Unfortunately, freeze drying and vacuum dehydration are also out of reach for most hobbyists. Keep in mind that this is my target audience. There may be an theoretical ideal extraction method that is accessible in a full lab environment that falls outside the domain of the average hobbyist.

I do understand that powdering and aqueous solutions are necessary for the testing steps and to get better homogenized results. If all other variables are limited, side by side results are still quite convincing.

The results on that graph do show small changes in aqueous solutions for the first 30 minutes. My results show that the percent change over longer duration is not linear. And 30 minutes is not nearly sufficient to dry mushrooms, so may not be relevant to a discussion of most effective dehydration methods. Given that potency loss is not linear but is a curve, we cannot determine whether one temperature is more effective over the required drying time based on the single data point on a different state (in solution vs whole fruit) provided in that paper. Plotting multiple curves may show that the lines cross at some point.

My testing only uses Qtests which show an accuracy within 10% by eye and within 5% by colorimeter when compared to HPLC for general use case. I concede that I wouldn't represent the mg/g readings as definitive. I do believe that tests done side by side with careful attention to limiting variables can be compared to each other, though. Especially if the differences are repeatable and fall outside of the stated margin of error.

Many of the other references cited simply make the recommendation to dry at lower temperatures, presumably based on prior art, but do not actually compare the effects of drying temperatures.

I am not even disagreeing with most of the citations. I just do not think that any show evidence of comparing drying methods to determine the most effective temperature for drying. That is the only hill that I am willing to die on at this point. If there is one I would love to see it. Some state that this is the case, but none of the ones cited appear to show that a comparison was actually undertaken.

I will be willing to make a bolder claim about which is actually the preferable method after some comparisons. I do not have sufficient fresh materials available right now. I can do a similar test to the last one and make a tea extraction using the vacuum chamber method and then drying multiple samples down to crystals using half at room temperature (dehydrator with heating element disabled; ~25c here) and another at full heat drying setting (74c). Those results will give an indication for optimal results for aqueous solution. Then I will do the same for fresh fruits when they are next available.

I honestly do not know which will prove more effective. It is easily possible that room temperature drying will be optimal. I simply have not seen convincing evidence that this has been determined base on anything I have read to date. Everything seems to address something approximately adjacent to the question or simply makes recommendations based on extrapolation, theory, or "common knowledge".

Call me a Missouri Mule, but I want direct evidence. Show me, or let me do it myself.

Again, I appreciate the engagement and promise to share my results no matter what they show. I know that heated dehydration is effective from experience, but I can't say if it is the MOST effective method or even if there is a significant difference between them, ... YET. I am traveling this week, so will have to do the next round of experiments in a week or more.

This paper from my source list looked into drying temperatures and potency.

Forensic analysis of hallucinogenic mushrooms and khat (Catha edulisForsk) using cation-exchange liquid chromatography.

Here's the chart from the paper of their findings.

Bear in mind that their 60C sample was likely dried in a lab oven with little or no airflow but you see what elevated temperatures do to potency compared to room temperature dried fruits.

There's a lot going on during the drying process that can affect the final potency of the dried fruits.

Firstly, the integrity of the outer skin as a barrier to oxygen entering the internal tissues. Mushroom skin forms a protective layer which stops external oxygen entering the internal tissues of the fruits. It also forms a waterproof barrier which means that when mushrooms dry naturally they dry from the inside out and most of their water content evaporates from the gills. When mushrooms dry naturally the integrity of the skin is maintained for longer as the fruits naturally divert water to the skin layer to prevent desiccation during periods of low humidity.

Dehydrator drying dries the fruits from the outside in and the integrity of the skin is the first thing to be affected which allows oxygen to penetrate into the internal tissues of the fruits where the alkaloids are stored. And of course it's not just oxygen. It's hot oxygen at pressure being blown at the fruits. And heat increases the chemical reactions like oxidisation that degrade potency.

Secondly, enzyme activity. PsiK, one of the synthesis enzymes has a dual function as a repair enzyme which rephosphorylates any unreacted psilocin produced in the fruits back into psilocybin and does so at three times the speed of the blueing reaction. So in essence it acts like a chemical brake to stop the blueing reaction from spreading through the whole fruit after injury. So PsiK protects psilocybin from degrading during the drying process as long as the enzymes are still functional and the fruits are metabolically active.

Enzymes are thermally degraded and become non functional at temperatures over 50C so with elevated temperature drying as soon as the fruits reach 50C PsiK's repair function stops working and potency loss increases as psilocybin degrades into psilocin and psilocin degrades into non active oligomers.

Interestingly, going back to the chart above you may have noticed that the room temperature dried fruits were more potent than the fresh calculated as dry weight figure which suggests that the Psi enzymes are still active and manufacturing psilocybin after the fruits have been harvested in low temperature dried fruits.

Then there's the effect of the crystallisation temperature on the stability of psilocybin. Psilocybin exhibits polymorphism, meaning it can crystallize in multiple structural forms, each with different structures which have different solubility, stability and melting points. Room temperature vacuum dried fruits result in the most stable form of psilocybin Polymorph A but elevated temperatures during crystallisation form Polymorph H which is an unstable form of psilocybin with a much shorter shelf life.

So not only are you reducing potency with elevated temperature drying and extraction processing what's left is unstable and has a much shorter shelf life. Not ideal for products like drinks, chocolate bars or gummies.

There's a fascinating VR video from the guy who co-authored the polymorph paper explaining the different crystal structures of psilocybin and how they form here.

1 gram of pure psilocybin will dissolve in 370ml of water so you could reduce the amount of water you're using which would reduce your evaporation time. That works out at about 10ml water per 25mg dose.

I'm pretty sure that psilocybin is so water soluble you get most of it into solution on the first pull too, so you probably don't need the additional extractions.

That looks like a lot of good information. I am running out the door but will give it a deeper dive as soon as I can.

Table 1 (the first image) is startling in the difference between room temp and 60c. It seems incredible based on my own experience, but it also seem extremely easy to test and verify or refute. A factor of 5 to 1 in total actives is well withing the margin of error of the Miraculix Qtests. So I will confirm or deny as soon as I am able and share the info here.

Maybe it is the movement of large volumes of air that makes the difference. I have seen that air fryers with a "dehydrator" function can destroy mushroom potency when run at similar temperatures to purpose built food dehydrators because they do not have sufficient air flow. Both from personal experience and in multiple reports of other users. Maybe baking vs dehydrating is really the source of all of the controversy. That would actually be pretty cool to discover. It will be a very interesting set of experiments no matter the results.

I did some testing of extractions of dried materials in multiple decoctions earlier in this thread. I found that the dregs retained a little less than 50% potency after each pull. Therefore, using a rough approximation a single extraction got 50%, second decoction gave a total yield of 75%, the third approached 90%. Psilocybin may be very water soluble, but the dregs don't give it up without a fight.

Thanks for sharing.

Rev, what I don't understand is how you're getting as little as 3g's out of 25 by only using a french press. That thing is not really a filter at all, you should be getting much more residual ... By using a cheesecloth and then some tea filter papers I still got basically half the weight of original material :/ My mushrooms were quite powdery, so maybe yours aren't that much. I'm trying a coffee filter and a medium speed filter with a buchner funnel (5-20 um) next. You can't really call 25g's out of 50 tea.

I wonder, if it's an issue filtering the tea somewhere half way in the dehidration process, when precipate has already started forming. It would be easier to filter 2L instead of 6 for instance ...

Btw, I tried a different batch of shrooms with my "oven tek" and it was fairly potent. Same procedure as before.

Good to hear that you are getting some better results.

Two likely reasons that the yield was low from that test. I didn't powder the mush like I have in the past. I just hand shredded the dry materials. There was some powder, but it was not nearly as fine as in previous runs. The other reason may be more telling. I only used 25 grams of dry material this time. That left a much smaller amount of dregs in the bottom of the french press. I think that my press handle reached the end of its travel before the dregs were fully compressed, so I didn't do a  vary good job wringing out the last of the tea.

Since both samples got the exact same treatment, I believe that the comparison is still valid. However, I would not repeat using the french press with such a small amount of mushroom material. In past tests with better compression of the dregs the relative yields were far greater as you found.

For the next trial of heated and unheated dehydration of aqueous solution I will be sure to use a larger mass of starting materials. It will all be from one larger batch of vacuum tea divided into three parts for dehydration testing. Some samples in 60c dehydrator, some in unheated dehydrator and some without a dehydrator in the dark. Actually, lets split that into 4 batches and add one at 60c in that air fryer dehydrator I mentioned to approximate the lab oven to see if we can replicate the results from the table discussed above.

I wish I could start this today, but I am packing up to head out for a few days and will have to do it when I get back. Need to get the wifey some much deserved beach time for mother's day.

Hey Rev, I have a quick question about the vacuum and heat test. I asked a question earlier but didn’t get a reply, so I’ll rephrase it—why did you actually do this test? Were you trying to see if there was any heat loss, or was there another reason?

Have a nice rest with your wife!

Okay, now I know why you did that test

So, what do you think — is making mushroom tea by heating the mushrooms in water to boil, then pouring all into a container and vacuum it, wait 15 minutes, strain and repeat with the dregs, a good approach? Or is it overkill?

If there's something I don't understand and it's a silly question, you can tell me :notsureif:

This is starting to sound a lot like the LODO brewers over at the hombrew forum

Let the water boil for 15 minutes to get rid of O2 in the water before adding mushrooms, purge the jar with CO2, add the tea, purge with CO2 and then vacuum :muppet:

This might sound like a crazy tea connoisseur idea, but I’m actually just making regular tea using the method described in this thread, but adding a vacuum :fancycook:

1 Add shrooms to water (with lemon juice - optional)
2 Bring to boil
2 Remove from heat, shortly after pour all into the jar in which you will create a vacuum
3 Create a vacuum
4 Hot water causes high pressure, so for the first 5 minutes you need to pump a little more every 1min
5 Wait 15 min in total
6 Drain liquid into cup, squeeze dregs, then repeat process with them

I get the feeling that there will be a slightly better extraction — which would be hard to notice anyway — but hey, its fun and I've already bought vacuum device, so why not give it a try? :psychsplit:

P.S. I'm also sure Rev has thought about this, so I'm curious to know his thoughts about it

Hey Blizzloo

You found what put me on the path to compare vacuum to heat. I have not tried combining them yet, but that could be the direction that we are heading. I do like Kloakk's brewing suggestion that uses Dave Bennett lead on de-oxygenated water.

@Kloakk - is the CO2 purge just to remove the air from the top of the vessel? I was wondering if bubbling through the tea would drop the ph which could have some potential benefits. Now we are getting into some more complex equipment, but it could be an interesting method to test some day. I feel like there might be some home brewing in my future if I were more of a beer drinker. I became a whiskey man in my old age, but big brother doesn't much approve of home distilling around here.

I don't know that the following would work better without some testing but it feels like the direction we are heading.

1 boil water for 15 minutes to de-oxygenate
2 place mush in a heat resistant vacuum chamber
3 pour in boiled water
4 pull a vacuum
5 steep for 15 minutes under vacuum
6 filter tea and squeeze dregs
7 repeat steps 1 - 6 once or twice

For immediate use, I might use some citrus juice or acid, but for drying or gummies I would pass on that step. I suspect that limiting the conversion of psilocybin to psilocin is better for long term potency retention. Plus it makes dried tea sticky and difficult to get dry crystals.
I would not expect to need to pull any additional vacuum. As the tea cools during the steep the vapor should shrink creating a stronger vacuum. You can test this by putting a two piece lid on a heated jar and then opening it after it cools. It will draw in a lot of air because the cooled air inside the jar contracts quite a lot. That is one reason that we use filtered lids and/or do not tighten unmodified grain jar lids until they are cool.

Keep in mind that the above is just a guess until I can do some comparison testing. So don't take it as a recommendation, but more of an interesting line to pursue next.

I am going to try to do the dehydrating temperature comparison of solutions this week. I found a six pack of half gallon wide mouth mason jars locally cheaper than a single one from Amazon last week. So I think I have my new vacuum chambers.

Well now you got in my head and I think I could do a heated vacuum vs unheated vacuum trial of the liquid teas. Then combine the resulting teas to do the dehydrating temperature comparison. That would save some time and materials I suppose.

Re: Top Tea Prep method test per potency testing of dregs, teas, & extracts [Re: ReverendMyc]

I've just looked out the research I was talking about up thread where I was saying that the majority of the alkaloids are extracted into solution during the first extraction.

https://static1.squarespace.com/static/5ea31b767e4ae053d67bd0f5/t/66479b295b7fa05c6185bd15/1715968810779/RCL-2024-Extraction_Study.pdf

It's interesting to note here how longer exposures to the test temperatures led to an increased loss of potency as I've previously stated.

To me it seems clear that double or triple extractions with the same amount of water or solvent to recover small percentages of alkaloids and the resulting doubling or tripling of the evaporation time combined with the additional heat exposure is going to result in a lower potency end product than if you just evaporated the first extraction.

Just to add some more detail to what I discovered about de-oxygenated water putting the brakes on the blueing reaction regarding the last few posts, oxygen from the atmosphere takes a long time to dissolve back into de-oxygenated water. And although hot water absorbs oxygen faster than cold water the amount of dissolved oxygen hot water can absorb is much less than cold water.

So in terms of tea making, pouring or making the tea in a vacuum flask, filled to the brim and then stoppered would exclude atmospheric oxygen from dissolving back into the tea water and protect potency without any need to be messing around with vacuums or inert gasses.

Secondly, there seems to be a lot of confusion about what weak acids like lemon juice do to the alkaloids during an extraction. Contrary to community lore, lemon juice doesn't convert psilocybin into psilocin.

Weak acids make the alkaloids more soluble and increase their stability in solution. So soaking the powdered dried fruits in lemon juice to make a slurry prior to adding the water or using acidified water results in higher potency extractions that are much more stable than if you'd just used water. pH 4 is the optimum acidity to shoot for in extractions according to the published research.

You can convert the psilocybin into psilocin in an acidified solution but you have to heat it to increase the enzymatic and hydrolytic dephosphorylation of psilocybin. As I mentioned in a previous post, heating the solution at 90C for an hour led to a 17% conversion of psilocybin into psilocin in the research.

So if I was making tea based on my knowledge of the published research it would go like this.

De-oxygenate the tea water by rapid boiling.

Acidify your powdered fruits with neat lemon juice to make a slurry and leave for 10 to 15 minutes before adding the water or acidify the water and add to the powder in a vacuum flask.

Fill the flask to the brim and stopper tightly to exclude oxygen and leave for your required brewing time.

Pour and enjoy!

Similarly for making extracts with maximum psilocybin levels for stability.

De-oxygenate water by rapid boiling.

Pour into a heat proof bottle or lab flask and fill to the brim. Stopper tightly to exclude oxygen and allow to cool or refrigerate.

Acidify your powdered fruits with neat lemon juice to make a slurry and leave for 10 to 15 minutes before adding the cold water or acidify the water to pH 4.

Add some Vitamin C to reduce alkaloid degradation during the longer extraction time required for cold water extractions.

Pour into a vacuum flask to keep the solution cold or perform the extraction in a vessel that excludes light to reduce photo-degradation. Leave to extract for your desired time.

I'm not sold on the idea of making dry extracts due to the inevitable loss of potency that occurs during the long exposure to heat during the evaporation process and their instability during storage but as I've mentioned previously using a vacuum chamber or a rotary vacuum dryer to evaporate off the water at low temperatures rather than using heat would lead to a much more potent end product.

Steam also displaces oxygen so if you are evaporating with heat a very low simmer or heating in a pressure cooker with the lid on but without the weight would probably be the best way to do it rather than blowing hot oxygen at the solution in a dehydrator.

Gummies aren't great for stability either due to their permeable and translucent nature which allows light and oxygen to degrade potency during storage but that can be improved by using light/air proof packaging or dipping the gummies in chocolate to form a hard shell which would stop light and oxygen interacting with the alkaloids.

Being a recovering brewer, my head went right to where @Kloakk was with LODO.

To all the folks that understand the chemistry much better than I do.
If we pretend eliminating oxygen is possible, does that suggest a stable liquid product?
Or are there other reactions that would continue degrading potency, leading us back dehydration?

LODO uses Campden tablets (sodium or potassium metabisulfite) to scavenge oxygen that inevitably sneaks in.
Does anyone have a guess or better at what effects that would have on the chemical reactions going on in tea?

This paper looked into the stability of the alkaloids over time and found that they were stable in solution for two weeks (which was the length of the experiment, not when the the alkaloids completely degraded) if light was excluded and the solution was refrigerated.

Investigation into the temporal stability of aqueous standard solutions of psilocin and psilocybin using high performance liquid chromatography.

Bear in mind that it was written in 2005 when they didn't understand that psilocybin degrades into psilocin so the loss of potency they found at seven days and traces at fourteen days has to an artefact of their handling process rather than the chemical properties of the alkaloids.

I filtered the tea through a medium speed filter and at the end got about a 45% extraction. I'm guessing this is it in regards to a water based extraction. Any idea how we could reduce this? LLM's are recommending me different methods, the best one might be alcohol precipitation:
- reduce the liquid to syrup-like consistency, still pourable
- cool down the liquid to 4C
- add 3–4 volumes of cold alcohol to 1 volume of aqueous extract
- wait a couple of hours for the precipate to form
- decant, filter and evaporate ...

Supposedly this should remove; polysaccharides, high molecular weight carbohydrates, some proteins, some pigments and mineral salts but amino acids and small sugars would remain ... I am a total noob when it comes to chemistry so somebody please comment if this would work or if there are better, cheaper options. One would need a vacuum pump and a buchner funnel for this and alcohol also isn't cheap; I'm figuring something around 1L of alcohol for 100g's of dried shrooms. Although on could capture the alcohol with a vacuum pump and a cold trap but that's a further investment and a complication.

Also, the french press option, seemed to serve you well as the final extract was pretty low in weight ...

Regarding just plain old dehydrated tea, optimizing the reduction process and forming crystals, I found this to be an easy and effective way, especially when it comes to the final step of forming crystals.

- Set the oven to 75C, partially open the oven door
- Pour the liquid in deep oven trays and reduce down to syrup consistency
- Pour the syrup in to a non stick pan.
- When the syrup has hardened, but is still a bit soft, remove the pan from the oven and carefully remove the soft extract. It's easier to remove if you wait a bit for it to cool down. Also, the thicker the layer the less chances of it breaking up.

- Place the extract on some baking paper in the pan and back to the oven.

- When the extract is mostly done, you can break it up with a pestle and mortal and put it back in the oven to optimize the final step of the drying process.

- Remove the dried extract from the oven and grind it down to fine powder.

From the point of starting the reduction process, this takes app 5 to 6 hours. It's quick because the metal oven trays heat up pretty well and the surface area is great; you can pour the tea in as much trays as your oven supports ... The negative is that you have to present for the crucial step of removing the extract from the pan.
