@SupaThaRipper you should post a link to this in Tri's TekDeck he is trying to gather a list of newer write ups / teks that are useful and a little more recent than some of the older lists that are still floating around.

realised that the bacteria’s spread is highly dependent on gravity

Can you elaborate on that? when I searched for "effect of gravity on bacteria growth" I only get studies related to space and really high/low gravity/acceleration.

Here is a photo of some bacteria on spores:

I don't think gravity is a relevant factor in how CS works. I also searched the publication for the term gravity it doesn't show up.

But in the abstract it says this:

Quote:
A handy method to remove bacterial contamination from fungal cultures said:
we developed a new method for bacterial removal from fungi cultured on solid medium, the Cabin-Sequestering (CS) method, based on the different culture characteristics between fungi and bacteria. [...] After 7–10 days of culturing, fungal hyphae grow out along the edge of the coverslip; however, the contaminating bacteria cannot pass through the space formed between the medium and the coverslip and, thus, remain in the cabin. The newly grown fungal hyphae around the coverslip are re-inoculated into fresh culture plates, where they form bacteria-free fungal colonies.

So its the agar-agar walls of the cabin that are physically blocking the bacteria from riding along on the myc as the myc penetrates the agar. If I'm understanding the paper correctly.

My assumptions here are that myc is really good at penetrating things since it helps trees share nutrients in the forest etc which is how it gets through the sealed agar cabin, hence sequestering the bacteria in the cabin.

Please anyone feel free to correct me if there's a flaw in my logic here, just trying to learn and spread the good word.

edit: I just scanned the paper a tiny bit more and in section 3.1 found the following

Quote:
paper said:
The M. oryzae hyphae can penetrate into the surrounding medium and bypass the coverslip to grow and form new colonies around it. In contrast, the bacteria are restricted by the solid medium and the coverslip, and, thus, they are retained in the cabins under the coverslip. The fungal hyphae around the coverslip are reinoculated onto new CM plates.

edit 2: I've since found out that the coverslips they refer to in the paper are actual coverslips and the demo video from the mushroom festival guy (Paul G) isn't actually following the paper by using agar for the roofs.

The paper calls out that they think coverslips are easier to work with than an agar roof, which is probably true. But I don't think using an agar roof would change the effectiveness of the technique.

MycCycle says that using agar for the roof is just sandwiching which I slightly disagree with since you're still carving out a cabin, but it is pretty similar to sandwiching.

Is there a way to separate the fungi i like from molds and other contaminants besides bacteria.

I'm thinking of using Pristine BRF puck tek, cabin sequestering and water agar for cleaning tools. I don't have a tool in my arsenal for cleaning contaminants other than bacteria though.

I'd say this reply from the previous page answers that question:

Will update later but also testing it for cloning, small mushrooms are always a pain to clone. I’ve taken 4 samples of whole stipe sections from zapotecorum and placed entire sections in the cabin. Samples of clone tissue were handled with bare hands and cut with non sanitized scissors eg handled in a very dirty way without regard. They were placed within the cabins with sterilized tweezers however, just trying to convey I took no care to ensure tissue samples were clean in any regard, rather tried to make them dirtier. All samples are now starting to show initial faint threads emerging out the cabin, no bacteria can be seen yet. Will update with pictures once things are more visible for pictures.

Double post, just wanted this one to be a stand alone.

Nice work, Supa. I'm looking forward to seeing the final results re pasteurization of spores.

In addition to that. Using cold temperatures is a good way to do so. I've been storing plates that I know will contam with mold in my cold fruiting chamber at 60; degrees and have been having literally jaw dropping results. ..

I have a lot of updating and revising to do on this thread. Hopefully can squeeze some time in this weekend. Have some techniques that I've been getting some results with for reviving old spores as well. Need to make new agar video with me actually venting the PC.

And yes Stipe, I need to add your spore pasteurization in here as well. I have yet to restart that sadly

Keep up the good work, looking forward to it. You've completed above average amount of work here, with plenty more to go. No rush, we're all

This sequestering method is producing stellar results while simultaneously answering the question regarding comingled mold/bacterial infections.

Sorry, only just saw this and thought it best to clarify, in all other cases CS worked in halting bacteria except for whatever was growing on these wild fruits, I’d find the bacteria escaping the cabin at the same time as the myc and growing in thick viscous blobs extremely fast filling a plate within days, by angling the plate the growth and spread of bacteria upwards was slowed by gravity giving the myc a better advantage to grow through beforehand.
While many bacteria can move in their own limited way their most common form of growth is simply fission, exponential growth over a food source, that growth is not unlike water spreading over a surface from a central point and gravity will always dictate movement to certain areas quicker than others, I simply used that to my advantage

Thanks for this thread Pheno!
For best results should I be sampling from the top of the roof?

Also, any chance you could fix those youtube links in the OP? Re-upload maybe if they were your vids?
