I don't know why I've never even considered using a thin glass pane and cutting it to the size of a microscope slide.

When I finally dip my toes into doing some CS I will follow your lead good sir.

I used microscope slides, but I cut them in half, and thanks for the kind words, but I am only a follower with the glass roof, my man MycCycle is the one who deserves the credit :awesomenod:

Oh man... I can't believe I forgot I was in this mass grow. Forgot about the thread but not about my cultures. Here's what I've got right now.

Pan Cyanescens

I have a lot of plates and have put some to poo-cakes and they are growing fast. I also have some on oats as well.


P. Cubensis ~ JFxPE

I have two shoeboxes. One created on Feb 6 and the other on Feb 8.


I have lots of grain in quarts going too...


P. Cubensis ~ KSSS

I also have 2 shoeboxes of this variety made on the 6th of Feb.


I have a lot of grain quarts for this too.


...and in single pints as well...


And I have them all tucked into a 110 qt. tub.


Leucistic Koh Samui

This is still colonizing and going well.


I also went overboard and have these cloned and/or started.

instant fail CS dirty semp myc, transferring from mold is hard if not impossible I think? trying again anyway, plus couple new plates from pucks.

Enmeshed mold is one of the harder things I've ever tried to transfer away from. Try switching up your nutrients in your agar and going from agar to brf puck then back to agar again.

Then if it's still there, try Josek Poking from agar to brf puck.

to get it strait, I think you mentioned this before but in the recent CS paper they use slipcovers, but in the video presentation of the paper by Paul Grzybowski he talks about sandwiching which uses an agar cover and plug on top of that, but when talking about the new technique he says at one point 'agar slipcover' and the meaning is perhaps not quite clear, but he must mean slipcover, as in the paper. he's also of course just talking about bacterial contamination, not mold, I think. I guess usually just tossing your mold plates is the smart move :lol:

February 15: A bunch of updating, condensed.

Soooo the GT plates didn't recover. Hardly need to speculate on why. BUT, today's update isn't about dropping out or weeping about. It's about...

LAGM 2025 (2), Electric Boogaloo!



Whipped up 12 no-pours yesterday. Swabbed a small bit of a kindly gifted print of Tico (ty Ax! :heart:) and gently rolled the swab to and fro in a zigzag upon 2 fresh plates. Also made a backup BRF puck with the spore-laden swab head.

side note this batch of plates is by far my best looking, even if they are a touch condensation-heavy. Even, pretty color... So a small victory! Huge props to Throwaway for the no-pour tek and Milkboy for the French Press PDA tek :smile:

I realized today that I can use the sealing bar on my vacuum sealer to gusset the edges of ziplocks to make them form nice cube edges for Fahtloc bags.

Another Tico jar was spawned.

Yeah, the way I see it the concept is misunderstood by Grzybowski and many others.
I cannot remember exactly what he said in the video and I do not want to disregard his competence.
However, in the presentation he shows a video where they use agar coverslips. He also states that it's agar coverslips they use.

The issue is that when you watch his video first, you're biased to think it's an agar coverslip that they mention in the paper.
Giving it credibility.
If you go the other way around, from the perspective of having a clear understanding of coverslips being those glass slides, you'll think "why is that guy using agar coverslips?".

The method is aimed to clean bacterial contamination. The kind of bacteria that is hard to spot and hard to get rid of.
To remove mold and separate from other funghi, isolation and transfers is the way to go.

As soon as you tightly plug agar into agar, there is nothing stopping the bacterial movement. No gap for it to overcome.

I also understand it so that the point of the coverslip is to force 2-dimentional movement of the mycelium for it to spread faster, instead of growing upwards.
This gives it a little boost to get away from the contamination. Which makes a lot of sense.

Some species have a tendency to grow straight upwards, touching lid. The point of the coverslip is to prevent that.


(Edit: I am in no way equipped to conclude with or state for certain that it's wrong. Neither have I done diligent testing or comparisons. I know I come of strong with my claims, but they're just that. Claims. I have a critical mind which is why I have engaged in this discussion with the hope that others are willing to try the different methods with an open mind so that we together can find the best, easiest, most efficient and reliable methods to cultivate full canopies of clean, healthy and robust mushrooms.)
