When isolating a strain, what exactly are you narrowing down? What does sectoring down to a monoculture accomplish? Does it ensure that every fruit will look the same? Or does it ensure that every flush will look the same? I’m trying to understand, what exactly is being honed in on.

Does anyone else get seriously down spirited when they've run out of agar work to do for awhile?
A whole mess of transfers from my MS plates today and now nothing else to do until they're all ready to transfer again.

I'm feeling like going on a print/swab spree just to give myself more shit to do this winter.

The idea is to hone in on desirable characteristics. Potency, colonizing speeds ..ect. The only issue is its impossible to know what you've got until you grow it out. So when you 'narrow down' you are going blind.

Imagine doing 60 transfers from ms and isolating something that doesnt fruit. That's why most people clone. But either cloning or isolating the idea is to have a consistent performing culture.

When times get like this I go through my plates and pick out all the fully colonized ones.
Wash, soak, sterilize grain.
Gather grain water
Drop all my fully colonized plates onto grain
Make grain water agar, replace plates used for grain.
Usually the plates I'm picking from are just transfer plates but you never know the kinds of genetics you've got until they've been grown.

Hey guys!
So I'm trying to revive this old bunk print of spores i had, they are Argentina. What do you guys think, is this Myc? I did 2 xfers to clean the myc of that black shit.

So what your saying is that unless I happen upon a massive potent fast fruiting cube in one of my ms trays, isolating a mono will be less than ideal.
Also, if I were to take a print from that isolated superfruit, put some on a plate, then xfer that to a mono, would it match the previous genetics identically?

Hey Adreno, if you take a print from a fruit you start from ms all over i think, you will get a random set of genes from that spore even though it's a print from a nice fruit.
Cloning is when you open a fruit and grab a piece of tissue from the mush and put it in agar. That way you know that the myc from that clone will perform kind of like the fruit you cloned.

Thank you guys. My mind is here now: Why mess with agar pins when you're uncertain of the pinset/ overall performance when you can clone from fruits that are already showing pinning performance? Are you mostly using agar pins from plates that are pretty isolated? Sorry curious.

Cuz plates are not the optimal environment so myc that fruits there should thrive in a tub.

Plus you never really know what you’ll get with a clone no matter where it comes from. I’ve cloned a big solitary fruit and gotten a crazy dense forest of three inch mushrooms.

I'm just doing it to do it.

the same reasons mushhead states... but really until i am cloning from select fruits its less likely to lead to a great culture.

but doing agar transfers is just work now.  having these little clones to fuck with is just fun. Definitely fun to watch.

I've also started a serial dilution project with AA+ . I did 6 1/10th dilutions from half a small spore print, so if there were a million spores per cc in the original right off the print (no way for me to quantify without a real microscope) there should be only 1 spore per cc in the 1/10^6 dilution. I could be getting the math wrong, i am just removing a zero with each dilution.



I'll probably plate 1cc of dilution ^2 thru ^5 and see if i can isolate some haploids.  If that is acheived, let's say finding 4-10 different ones.  I'd then hit each one to all the others at that point. Though AA  would be a good variety to cross with others since there is the potential for the inherited albino trait.

Would be cool to isolate a haploid AA+ that consistently forces albinoism on its mate.

Lotta work to get there, if at all.

Hello eveveryone,
I was told this was the place to come for agar advice. These were inoculated using a spore syringe and a loop. I have no idea what they're meant to look like, these are my first plates, but to me they look weird. They were made and inoculated on 1st December. What do you guys think?

Hey everyone! I finally got around to agar and am about to transfer for the first time! How do these look? I understand the quality is bad, it was difficult to get clear pictures without reflections. Everything pictured should be mycelium besides the Tampanensis plates, I could use advice on those. Could they be sclerotia?
