If you want to test your agar, try leaving some dishes untouched after PC and see if they grow anything. If they do, then something is wrong in your sterilization or the containers are faulty. But opening them up, coughing on them, etc doesn't really tell you anything about your agar.

You could also open some up as if you were going to transfer myc, but instead don't do anything with them. Just go through the same steps without actually transferring. If you get mold growth, that tells you something about your technique.

Yeah, that's what I am gonna do next time I do transfers. Good idea.

With regard to simply leaving plates untouched, that's what I do anyway, same with grain jars. I leave at least one of each batch untouched and have never seen any contam on those. So the preparation process is fine at least.

Found this helpful. This suggests to treat the translucent growth the same as you would white myc growth.

Hopefully you haven't answered this already, but how old are those plates? That sort of growth is exactly how even healthy transfers grow on my mea plates when I store them for too long (months) before using them.

Sonic's "I'll buy a pressure cooker someday" Instant Pot times and settings:
  • Popping Corn Rapid Re-hydration - Half fill all containers with corn. Empty into Instant Pot. Cover with water. Run 30 minutes on PC. Release, remove, dry.
  • Wheat/Rye Berry Soak Re-hydration - Half fill all containers with grain. Empty in pot with water. Soak for 6-12 hours, air dry until surface is completely dry.
  • Grain Jar Sterilization - Run 2 - 2.5 hrs on PC
  • PastyPlates, MEA Formulation- Run 45 minutes on PC
  • LME Liquid Culture - Run 45 minutes on PC

Nah, they aren't old at all. just few weeks. I have not really figured out why this translucent growth shows up and I am having trouble believing it is something other than mycelium, because it also changes to strong rhizomorphic growth on some plates after a while. well, maybe it could be escaping something, but I am very unsure about this. Have recently seen translucent mycelium changing to strong growth after changing to MEYA from MEA, but it started out translucent as well and 1-2 week in it took off.

This is very helpful. It's the first time to see this is happening to the pros, too. Man, am I glad I am not too stupid to pull this off :smile:

I guess we still don't know, what it is that's going on, but at least it ends all good.
