my spore wont take to any of the rye berries i use I NEED HELP

I've inoculated and grown lots of colonies until last summer.
Now, All substrate I buy leads to no growth at all. After some research I thought that my rye grain supplier changed their grains to rye berries grown with "fungicide". After trying a different local store and organic rye berries from amazon I'm having the same problem.

Its been a frustrating year of trial and error with no success. I've tried different methods of hydrating the rye before sterilization , and adding coffee to the mix and still no success.

I ordered some grow bags of pre sterilized rye grain and they colonized. its expensive to keep ordering them, and a huge hassle compared to cloning a whole bunch of jars.

I can post pictures of the jars after 3 weeks of incubation
but they just look like jars of pressure cooked rye seeds

I've been discouraged from this whole experience because until last summer I had been growing colonies for 3 years with out any issues.
Now I hit a brick wall and they wont start any mycellium growth when I ha vent done anything differently from all my previous success.

Please help me with any information, or advice, I cant be the only one who has been through this

I can only suggest to start doing agar work.

Shouldn't be putting spores to grain anyways, theyre probably dirty and contaminating the grain

I am scared of having the same issue as OP. Is agar more robust than injecting spores into a bag? Also do you drop the whole dish into the corn/brf/etc or can you inoculate multiple times from the same dish?

Its cleaner to get to take the guess work out of knowing its dirty as long as your technique of agar to grain is clean. NO, you don't add to sub, you add to your grain and spawn from that.

Spores are inherently dirty and the bacteria/mold spores will germinate and take over way before the mushroom spores even germinate, success rate is mabye about 50/50 and even then your "success" is a few meager flushes at best

While as a clean culture on agar put to grains your success rate is almost entirely dependant on your sterile technique and will produce beautiful flushes compared to bacterial spawn

Thats why we use agar, is to get a clean culture and get the best flushes possible

I have put spores to grain before, several times.  It often works, sometimes it goes rotten.

All these posts telling OP to use agar are beside the point.  The spores won't germinate.  And he's not reporting contamination. 

But then OP buys pre-sterilized rye and it works.

Good grief.  What's going on?  Im thinking it must be dead spores and the pre sterilized rye episode was a coincidence.

I print in a clean kitchen with sterilized containers and tinfoil, I never have dirty syringes I use a control jar before mass inoculations and sterilized water to refill and reuse the syringe. Cross contamination can occur but its yet to happen to me. But what works for me might not work for OP so anyways back to the question.

If your spores fail to germinate on one substrate median then try another. I had the exact same problem as you OP a little while ago, my spore syringe was made properly with fairly fresh spores from a previous grow but when I inoculated grains from various different sources the spores would not germinate....I thought it was fungicides but never figured it out because I switched from wbs and rye to popcorn....the popcorn worked.

That's the only thing i can suggest is to buy it from a grocery store so you know its food
Grade and if you get germination then you'll know if they're treating your whole grains with fungicides or pesticides since popcorn is food grade it won't contain any of that.

Keep the thread updated if you do try it out since I'm curious to know myself about the spore phenomenon.

Its way easier than injecting spores into a bag ( unless you buy everything pre made and sterilized). And you have wayyyy better chance at success and freedom of developing superior culture.
Yes you can drop an entire dish into corn, brf, oats or for me wbs. Or just wedges from a plate. I use max 1 plate into 4 quarts of grain but I'm sure you can do way more.
Get into it bruh. It's so cheap and non time consuming.

When I decide to do a MS injection I choose to use BRF cakes. If I want bulk, I just wait until it’s fully colonized and use a cheese grater wiped with alcohol and shred them in the the bulk substrate.

What Rye prep TEK are you using?

lol seemed like you were the only person to really understand my post.

the whole thread turned into a just use agar, when a dirty syringe was nt the issue. and never had any containment's

are you willing to take the time and try to help me?
through some research I thought maybe it was "fungicide" in the rye  but all the places i buy it from my self doesnt work
so i felt like that might be a little far fetched

do you know if something with my pressure cooker may be messing it up ? because thats all i can think of that im doing before inoculation

you inject spores into rye grain jars YOU made and nothing happens? no growth no mold?

you use the same spores into vendor bags and you grow shrooms?

vent the pc for  20min but if the cycle was to blame the jars would mold fast af

Have you been using spores that are all from the same source all this time?  I want to believe it's the spores. 

But if it's not, I guess it must be the rye or the water you are putting in the rye.

Are you shaking the syringe really hard before each inoculation? It's something I've forgotten and it makes a huge difference - easily testable with agar.

Everyone understood just fine. If you go to agar first you can tell if you don't have contamination and Also tell if you have good germination. Yea they worked in the bags but not your grain. New growers have a hard time noticing contamination. Perhaps those bags were bad but looked pretty decent to you. If the spores worked on n agar youd know your grain prep was bad. Its pretty hard to do anything but shoot in the dark if you didn't do proper diagnostics first like using agar to germinate.
