agar contaminated?

hi,

this is one of the very first agars which I made at all as a newbie.



is it contaminated? should I dump completely?
or would it make sense to save some parts of it by making another agar?

one agar shows nothing, no growth at all, this one above looks suspicious for me, the third one has only white stuff growing in it, all made with Copelandia Hawaii spore syringe.

the other three agars were made with Golden Teacher, by all three looking fine.

agar was made of 350 ml water, 7 g LME and 7 g agaragar, then poured into 6 petri dishes(, but not all of the agar was required).

thanks for any advice!

hello, me too am new to this cultural world, yes it is a green mold, I advise you to isolate your mycelium (white part) because it's healthy by avoiding taking the ends of the green mold, and deposit in a new petri dish. the abbreviation LME means what (complete word)?

Light Malt Extract.

Take a transfer or two from the growth just above and to the right of the "green mold". Do this in a Still Air box with a flame sterilized scalpel. I'd take a least one, maybe two or three, transfers from the outer edges of the three lobes of that growth. The center lobe (sector) looks the most promising to me. It may be dirty still but that is what agar is for.

I can hardly wait to start using agar, myself, but I travel a lot, and sometimes without notice, for days on end.

I just took three samples as you instructed (if I understood correctly) and made three further agars:



do I have to isolate until I have a monoculture in order to inoculate grains?
so maybe do a another isolation from these (if they are clean and successful)?
how much agar sample is sufficient for grain inoculation?

You need a monoculture in the sense that it must be only cubensis myc in the dish. No yeast, bacteria, imperfect fungi...... Just magic. The term is used in other ways but you definitely don't need a monoculture in the single sector isolate sense.

Looks like you understood perfectly.

You can put as much or as little in a jar as you want. The more you put in the faster it'll grow. I don't usually do under an eighth of a plate though.

Transfer till you have a single, clean, symmetrical culture. You don't want any satellite colonies.

Usually a good rule of thumb is to take at least 3 transfers from spores. I have used my T2 plates if I thought they were clean enough. You will just have to get an eye for what a clean culture looks like.

When I go a2g, I allow my plates to grow out till almost the edges (not all the way to edge tho). Then I use a whole plate, cut in a tic-tac-toe pattern or like a pizza is cut (just depends on how I feel that day), to inoculate a at jars of hydrated grains. The reason I cut into pieces is it gives several inoculation points.

I don't know about "until symmetrical". I get your point but sectors often appear and are not symmetrical. Definitely no satellites though

Maybe uniform would be a better adjective then symmetrical. I look for even growth, from my original transfer, all the way around the plate.

hello, I try to grown agaricus bisporus but it takes me a lot of time, I would like to know if you too have this same problem ? i know for the culture of oyster mushrooms is faster, but I would like to have your opinion

I see what you mean. Yes I do get that growth. Just been shooting for what I described. 

Learned something today, did not know that was sectoring (the term) and yes it is not quite uniformed or symmetrical.

Love this hobby, you absolutely learn something every single day.

Look into some of THESE POSTS! There you have every time a 'trusted cultivator' mentioned it in a post.

pro tip: this was achieved by clicking on "search posts", Typing in the desired term, clicking the "trusted Cultivator" tab and clicking search. It's a very useful aspect of this site. Also, to further refine the search, you could designate how old or new you want the post to be. Also, this is a edible or gourmet mushroom so you'll want to search the gourmet forum for more info on it. Peace
