2 weeks since inoculation very little growth.

Hey i nocced up almost 20 pf tek jars and am seeing very little growth after 2 whole weeks.

I have also 20 qt jars that arent showing any growth after a week...

Is it the vendor syringes to blame here considering everything else was done correctly?

And how long until theyre considered garbage?

You didn’t really include any information as to how you did anything, so I wouldn’t go straight to blaming the syringe. What procedures and techniques did you use, etc? Also Syringe to Grain is a shot in the dark if you ask most people around here, everyone is gonna suggest to learn agar and I would take the advice and research and study all you can on it, I’m new here as well and just got all my supplies to get Agar going. It’s 120% worth looking into if you’re serious about the hobby.

it can be like that sometimes.  A week to germinate isn't that unusual.  I have had two prints that never germinated, I waited over a month before I threw the plates out.  I have had one print that was too slow, I can't say its the vendors fault, just weak genetics, maybe it would have done better on a different agar.  Just be patient One of the slowest clones I work with ends up putting out a lot of really nice fruits.

I did franks 12 steps to shrooms basically.

Ms inoc through polyfill using 5-10 drops which i kinda thought was very little (considering 10 ccs will do 10 pf tek jars) i used about 1 cc for 7 jars.

Last night i actually had a thought that maybe i wasnt squirting enough out after flame sterilizing ... as in i was inoculating using sterile liquid but i dont think this was the case.

Yeah i didnt mean to blame the vendor i meant to blame the genes of whats in the syringe.

And it does seem like that doesnt it... the slow growers sometimes put out some big ones.

Usually proper temperature needs to be held. Now if jars remain normal without any discoloration then sterility had no role to play. The substrate prep also is key to rapid colonization, too little or wet slows things down inviting contamination.

Telling people to toss growing jars that are a little behind  after a couple weeks is beyond asinine. Multispore can take up to 3 weeks to germinate on grain. OP is ahead of the game.
I'm beginning to think this site has a lot of insecure people lurking that purposefully give bad advice and misinformation. That and some egomaniacal "shroom gods" that like to hear themselves talk and are one track and closeminded as they come.

You would toss 40 jars after 2 weeks, 20 after a week? That's about 8 hours worth of prep that you would throw in the trash in the name of being impatient in this hobby of patience.

OP, have patience. Give it a week then give us some pics.

You only used 1cc spread out across 7 jars? Or 1cc per jar?

We have an expert with a picture of a book  :bow2:  Where in the book does it say to toss it and move on? I don't see a section on inoculating grain with spore syringes. I might have missed it though... I am not an expert.

1 cc across 78 jars. like 5 drops per jar.


I am being patient. next time i will be inoculating against the glass like you do in pf tek so i can spot the germination easier.

Ill update this post when i can see some growth.

1 pf jar out of almost 20 is showing growth, so thats a good sign.

Exactly.

Would not be happy about the 4 90 minute pc cycles and all other prep work being wasted.

That seems like very little... But if you followed a tek and they say it's good, then rock on. I think there are a few thousand spores in a drop. And I also  read in that book quoted earlier that " for every one hundred spores, only an average of one to five germinate", and they need to find each other to mate. so that seems like a small amount for cakes... Everyone else seems to do fine with a small amount of spore solution though :shrug: I've been barely able to germinate on agar using just one inoculation loop worth of solution. Next time I'm gonna increase the dose.

Hi ...

I did some agar work and all of the plates i inoculated via syringe show bacterial growth from my "ecuador" and "amazon" syringes.

These are the same variety i used to inoculate the jars that arent growing.

Pretty sure all that work i did for nothing . look at it all .



The ones you see growing are the ones that are done with proven genetics.

Everything else, including 9 qts more of rye underneath not shown, and like 20 agar plates ... is expected to contam.

My next question would be-

Can i re- pressure cook all these grain jars and inoculate with agar?

or do i have to toss the 21 qt jars of rye berries? (please no)

I have re PCed grain jars.  It works, if they have a lot of bacteria showing you may want to toss, but in my opinion it would be better to pc and noc them up again with an agar wedge rather than toss all that prepped grain

What about moisture content? Could they have dried out too much too reuse?
