Started my hand at agar.  Did MEA.  These are my first pours.  I used a MS syringe.  I tried to inoculate 10 plates and only had one grow ANYTHING.  I think i did not let my needle cool enough after flame and only put one drop on each plate.  One drop seemed like a lot of liquid but i think i cooked most of it.  :foreheadslap:

here is my only survivor.  he looks pretty damn clean and isolated to me.  Is there value in transferring or should i just let him grow out and use for grain inoculation.  Seems like transferring him would just get it to the center of the plate.  I know it is t word for fluffy growth but i know that does not matter.

Looks good you need to make a transfer from the leading edge. You should make atleast 2 transfers to make sure you get a clean plate.

Ready to transfer, looks pretty uniform. You can take that out to ten or more plates if you like. Yeah at least a few transfers is best to ensure there are no hidden contaminants. Other than that the benefits are that you will be narrowing down the genetics, which is desirable, and you can turn that one plate into as many as you want. Then you can put 10 or 20 (+) agar wedges to grain, rather than just 1 or 2 from that plate.

Btw, isolate is usually used in terms of strains, probably not what you mean here.

Oh and when I do MS solution to agar, I like to let a drop out onto the floor of the SAB after flame sterilizing, to cool the needle.
