Have any of you guys ever put spores onto a dish and it germinate EVERYWHERE because of the condensation water and wateryness of the agar . Haha I got a few Transfers deep and it’s still just everywhere.

Hoob... avoid tilting the plate in the future: this will spread contamination like nobody's business... if the culture is clean, and your technique was sound both pouring and transferring, then there's no worry... if I'm reading correctly, this is a few transfers out from the germ plate, and has no spores present? Pour your plates cooler to avoid condensation: 117°F is said to be optimal

Just wanted to say that I did 20 plates of clones and transfers and didn’t get a single contam.  Can’t believe the clones came out clean. 

I’m getting good at this I believe.

I’m doing pasty plates. And yes I think I may have tipped  them when I was check them out
Yes I’m a few transfers in. Got the cleanest one and made 5 plates out of it. All looking good so far 
And hopefully going to grain as soon as I prep some

Reishi seems to be pretty easy (when you don't freeze it outside for 2 days mistaking the fruit for dry bubble) I was/will be growing it in a shoebox and treating it just like the cubes... I hear most oyster varieties are forgiving... really just opening the "outside of cubes" can myself, but I'll let you know... currently have reishi, bunapi (white shemeji), and king oysters on agar and about to shift research and focus in these directions

Forget treating gourmet/medicinal like cubes. As you have a hood now, you need to look into bags with sterilized sup’d wood substrate.  Oyster do well on pasteurized straw but it’s messy and bulky.

Well I just did my first ever transfer. When I took the myc from the one plate and put it to the other.. the piece of agar landed on the agar. Like the myc is facing up from the receiving dish's agar.

I'm assuming the myc will wrap around and start growing on the receiving dish, correct?

my first agar tubs PF classic going strong, made 2 sets of transfers but not looking to isolate yet, just nice and clean and i guess less variety than the germ tub.
I dropped two clean agar tubs i had transferred from into a 1L BRF jar and thats taking of nicely :laugh:

first transfer 5 days, took 2 chunks from each and that should do me for now 


used one of those tubs for blenderless LI earlier and inoculated some random tubs, about 4.5 pints together




havent been so lucky with AA+ syringe, but got one doing great and made transfers



definitely hope for these but i threw them to reuse the tub

Heres some contam showing up on an older tub :/ i already have transfers. looks different to the spunky white shit i've seen before.




Small orange blobs, i removed the largest one, but looks like i only succeeded in spreading it :L not a big loss but i was looking forward to just leaving a tub to see what goes down, will the myc eat it?

i am yet to put anything into my fruiting tub... which is why im not interested in isolating just yet haha

But when i do have fruits, would taking a tissue sample to agar grow out immediately as an isolate?

Those blobs are probably bacteria, you transfer away from contaminants, to new clean dishes, don't try to remove the.

Yes, taking a tissue sample from a fruit will get you some nice growth. Not sure if its a true isolate, but close enough for this work.

Vibe, the wedge doesn't care which way it faces, just on the surface of the new plate... I've noticed more even growth with the culture either face up or face down, and the center seems to get fluffy if it's facing any way but down... if the culture is rhizomorphic, I've found that lying the wedge on it's side can cause it to grow out tomentose on the side  opposite the culture face... still viable, as long as it's clean

Swissrolls, the orange was certainly bacterial colonies, and won't effect your culture without making contact, and only where contact is made/spreads... bacteria doesn't sporelate arially so no worries... I've even marked bacterial colonies in marker on the bottom of the plate, let the culture grow over it, avoid it taking transfers from the opposite side leading edge,  and got clean culture in one transfer... bacteria isn't that big of an issue on a 2d surface, neither is mold if you transfer before it sporelates too heavily... this is the number one reason we use agar...

alternatively, and not suggesting this as regular practice, but I have, in a low plate/no funds sitiation, successfully cut out bacterial colonies and fruited the plates they came from... trick is to cut a wedge with a hot scalpel, 1/4" or so all the way around it, and carefully scooping it out without disturbing it

My thing lately has been to try and remove a fraction of culture without transferring any of the agar... for no other reason than "just cuz:shrug:"

Also, a clone will typically still contain multiple strains performing together as one, often times still too numerous to even display sectoring, and is generally considered a monoculture... an isolate is a single set of genetics derived from isolation through many transfers: the smaller the transfers, the faster the isolation: though I wouldnt focus too heavily on isolation, as reported by RR,and my own experience,  the original cloned monoculture will outperform 11 of 12 isolates derived from it...

You should try King Oysters in a unmodified tub. Grows like coral and in large quantity.

Oh man, I just did agar transfers for 4 almost 5 hours straight. I don't know how y'all keep up! A week later they're all grown out again! Lol. I think I'm just doing too many varieties at a time. I need to cut back to like 3 or 5. They're so many I want to try tho! :lol: Its so hard to keep up :facepalm3:
