First grow in 12 years.

My how things have changed after 12 years lol!  Previously did a few PF Tek grows and casings in a PMP with some LC work.  Got the itch to try again and this time doing monotub.  I understand some of my stuff is outdated but I like old school.

Strain is MS Ecuador that I inoculated into a karo LC on the 4th.  Placed in a TiT incubator that is at 78F (made today as unable to control 68-75F swings throughout the day).  Currently decent growth in the LC and can't wait for my magnetic stirrer for next batch.

Will be using rye as my spawn.  I bought 4lbs presterilized rye that I will be comparing with my own quarts of rye/gypsum/coffee and 10lbs of manure based compost.

First monotub is a 30 quart that I want to  use the 4lbs of rye and 10lbs of compost with.  Measure and seems that will be about a 3.5" depth with about 9" head space.

Any pointers are appreciated.  I do plan on making SAB, getting into agar, and making my own substrate but wanted to start with poo plus baby steps.  I will eventually throw up some pics.

I used the self healing lid tek with SFD for my quart jars.  Very easy to follow and they look professional.  I saw Wal-Mart has plastic lids but not sure how well those hold up in pressure cookers.

You nocced up the LC directly from the spore syringe?

Hey man, good luck on your grow. A lot has changed even from when I first joined.

I just want to point out that no one makes LC directly from a spore syringe. The safe way is to go to agar first
and then noc up your LC from agar wedges. Heat sources are also generally frowned up because it provides a better
environment for bacteria that may be preset. I do LC and it does just fine at room temp (around 70F).

Also, I can tell you are old school :lol: No one calls them "casings" any more. Casing is specific to a casing
layer on a substrate only. Oh, skip the coffee because it will be more prone to contamination.

Always good to see someone return to the fold.

Spores directly to LC is going to be the issue with your grow. Who knows what's colonizing that LC if you noc it up directly with a syringe? Start an LC with a clean culture.

Yeah I've been reading up on agar and want to start doing that.  Didn't realize it was so easy to implement.  My last few grows were LC with spore syringe in open air and they did fine.  This time I used rubbing alcohol, flame needle, alcohol soaked sponge and a walk in closet I've cleaned out and bleached (beer closet to mushroom closet lol)  Will only knock up my bags before I knock up my other quarts too.  If that doesn't pay off it should give me enough time to gather more supplies.

Also cool that jars don't have to be incubated in the dark... the not looking was always the hardest part lol.

If you make an LC this way
Pastywhyte's EzLC Tek
than you can see what's growing in it and be sure it's clean before you noc up a bunch of grain with it.

I'm glad you haven't inoculated anything with that LC yet. I would abandon it.
Forget about LC until you are very well versed in sterile procedure , agar work, and grain to grain inoculations.
LC is an advanced technique and needs to start with agar, period.
Grains should only be inoculated via a clean culture on agar.


You can also ditch the tub in tub incubator! 78 is too high and will likely encourage bacteria. 68-75 with temp swings is just fine. You wouldn't need a heater unless your temps are below 60's, in which case a space heater in the room is the better way to go.

I would leave the coffee out of the grain. A little gypsum is fine but coffee always complicates things.

I would also really suggest using a brick of coir instead of manure. I would only use manure for Pan grows. For cubes, coir bricks are cheap, fast and simple to prep and require no pasteurizing. You can just put the brick in a 5 gallon bucket, add the amount of boiling water and cap. Let cool/mix and your good to go. It is very resistant to contamination. Anything with manure in it needs to be pasteurized, kept at a temp between 150-160 for one hour.
Almost all of us that grow Cubes have switched to straight coir substrate these days and it's been a huge improvement. You can buy coir bricks in the lizard section of any chain pet store

Thanks for all the suggestions.  Will leave the coffee out of those grains.  How much quarts is a normal brick of coir going to make? Trying to wrap my head around the measurement of my tubs.

As for the LC I decided to keep this one going and in the meantime order some agar and do the pastywhyte agar tek.  I will shoot up a couple with the LC and see if it contaminates.  I decided on LC before I started this grow... I remember my last few were using similar LC and I never contaminated until the 3rd or 4th flush which was probably due to other factors.  I do see the commonsense approach of just using agar first.  I have tons of rye available and time on my hands. So in 3 months I have no doubt my setup will be getting up to spec and multiple specimens to watch and experiment with.

Another question on coir as I am not sure if it is outdated or not.  Is manure going to give bigger fruits than coir?

About 7-9 varies a bit with brand. I get 9 pretty consistently from the ones I use.

No. Many of use get wall to wall canopy flushes with coir. Clean spawn, good genetics, dialed in surface conditions are what give great flushes.

Coir can be as little as 2 bux at hyroponic/gardening stores, at pet stores it can be like 4 or 5 , or you get 3 packs for cheaper.

I get closer to like 14 quarts. Its enough to do a full Monotub

So I've somewhat changed gears.  Had to order agar off internet but hope to have some plates made this week.  I want to use the LC I have now and see if I can get a good wedge I can transfer to another LC.  SAB should be done tomorrow all 100 quarts lol

I still would wait until you are very proficient in agar work, agar inoculations, G2G etc

But don't use the LC you have now to inoculate agar dude, if it's contaminated, which it is more likely to be than not, you won't likely be able to transfer away clean growth.

Just use spores to inoculate your agar and transfer clean myc from where it germinates

Spores to agar will work for me.  My worry is I bought syringes when I should have went for the prints but at this point I can make up to 18 dishes if needed.

Reading up while I wait.  I am more of a hands on person so stuff will start clicking more with trial and error.

Yea I prefer prints for agar inoculation but you can do a whole lot with what you have - you only should use a teeny drop to inoculate a plate and I only inoculate a couple plates to germinate a culture
